摘　　要：目的:观察海洛因戒断、复吸大鼠脑VTA、NAc内亮氨酸脑啡肽(leucine-enkephalin,Leu-Enk)和胆囊收缩素(cholecystokinin,CCK)的变化,探讨CCK和内源性阿片肽在海洛因戒断、脱毒和复吸过程中的作用及其相互关系。方法:正常雄性SD大鼠63只,随机分为实验组、盐水对照组和正常对照组。实验组大鼠又分海洛因戒断组、美沙酮脱毒组和海洛因复吸组。取实验各组及对照组大鼠脑组织,定位VTA和NAc,用免疫组化SABC法显示VTA,NAc区Leu-Enk,CCK阳性细胞并用图像分析法测定平均光密度值。结果:(1)Leu-Enk在实验各组大鼠VTA和NAc内的表达与对照组比较均显著降低(P<0.05)。(2)与对照组比较,海洛因戒断、复吸组大鼠VTA和NAc中CCK阳性细胞免疫反应增强,差异有显著性(P<0.05);经美沙酮脱毒后恢复正常。结论:海洛因戒断造成的机体应激及阿片类药物均抑制了VTA和NAc中Leu-Enk的分泌;CCK分泌增加,参与了对脑神经的保护。Objective： To investigate the effect of the heroin withdrawl and relapse on the expression of leucine- enkephalin （Leu-Enk） and cholecystokinin （CCK） in the ventral tegmental area （VTA） and nucleus accumbens （NAc） regions, and exploring the effects of CCK and endogenous opioid peptides during heroin withdrawl, methadone detoxication and heroin relapse period, and the relationship between Leu-Enk and CCK. Methods： Adulthood 63 male SD rats were randomly divided into three groups ： normal control group （ NCG）, saline control group （SCG） and experiment group （EG）. The experiment group was divided into： heroin withdrawl group （HWG）, methadone detoxication group （MDG） and heroin relapse group （HRG）. Brain tissues were detected after models were set up, VTA and NAc regions＇were be located. Immunohistochemical staining was performed to the mean density of Leu-Enk and CCK in the VTA and NAc regions. Results： （1）Leu-Enk-immunoreactive neurons expressed less Leu-Enk in the VTA and NAc regions from EG rats than that from SCG and NCG rats （ P 〈 0.05 ）. （2） The more CCK-immunoreactive neurons were observed in HWG and HRG compared with SCG and NCG（P 〈 0.05）. After methadone detoxication, the expression level of CCK became normal. Conclusion： That heroin withdrawl caused stress and opiates can inhibite the secretion of Leu-Enk in the VTAand NAc regions. More CCK secretion participated in protection of brain neurons.
