摘  要:目的探讨乙醇性痴呆大鼠海马区脑源性神经营养因子(brain-derived neurotrophic factor,BDNF)的表达变化及其与学习记忆功能损害的关系。方法 SD大鼠随机分为2组,每组6只,其中一组按体质量8mL/(kg·d)给予20%(体积分数)乙醇灌胃持续28d,建立乙醇性痴呆大鼠模型;另一组予等量生理盐水灌胃作为对照。观察两组大鼠Morris水迷宫行为(逃避潜伏期),采用HE染色方法观察海马组织形态改变,采用免疫组化检测大鼠海马区BDNF表达。结果乙醇组大鼠灌胃后逃避潜伏期时间(EL)〔(16.39±3.11)s〕较灌胃前〔(7.12±1.18)s〕明显延长(P<0.05);与生理盐水组灌胃后EL〔(5.76±0.86)s〕比较,乙醇组大鼠灌胃后EL明显延长(P<0.01),且海马形态结构明显破坏。免疫组化结果显示,与生理盐水组相比,乙醇组大鼠海马齿状回(DG)区(积分光密度值为111.65±14.44 vs.61.55±17.44)和CA1区(积分光密度值为88.98±9.15 vs.55.21±8.93)BDNF相对表达明显降低(均P<0.05)。结论乙醇可导致大鼠学习记忆功能及海马组织损伤,其机制可能与抑制海马BDNF表达有关。Objective To study the role of brain-derived neurotrophic factor (BDNF) in the mechanism of the ethanol-induced damage of learning and memory function in alcohol-associated dementia (AAD) rat model.Methods SD rats were selected randomly (6 rats in each group) and intragastricly administrated with ethanol (20%,8 ml/kg) or saline (as control) for 28 days.And animals were evaluated by observing Morris Maze behavior (escape latency,EL) and hippocampus morphology with HE staining.The expression of BDNF was detected by the method of immunohistochemistry.Results As compared with prior to ethanol administration [(7.12±1.18) s],the EL time of rats in the ethanol group was significantly extended after ethanol administration [(16.39±3.11) s,P〈0.05].As compared with the saline group [(5.76 ± 0.86) s],the EL time of rat receiving ethanol was apparently extended after ethanol administration [(16.39 ± 3.11) s,P〈 0.01)].And the result showed that the morphological structure was apparently damaged in rat hippocampus after ethanol-intragastric administration.As compared with the saline group,the expression of BDNF decreased significantly in the hippocampus (DG:111.65 ± 14.44 vs.61.55 ± 17.44; CA1:88.98 ± 9.15 vs.55.21 ± 8.93,P〈0.05) of rats treated with ethanol.Conclusions The damage of learning and memory function and hippocampus morphology of rats induced by ethanol is closely related with the inhibition effect of ethanol on BDNF expression in the hippocampus.
