摘 要:目的:评价壳聚糖温敏水凝胶与牙周膜细胞的生物相容性。方法:将牙周膜细胞接种于壳聚糖温敏水凝胶中固化后进行三维培养。通过MTT法、细胞周期分析,活/死细胞染色法检测牙周膜细胞与壳聚糖温敏水凝胶的生物相容性。结果:壳聚糖溶液加入β-磷酸甘油钠后具有温敏特性,在生理温度下固化形成凝胶。壳聚糖水凝胶提取液对细胞增殖没有影响,并可提高细胞S期的比例。牙周膜细胞在壳聚糖水凝胶中三维培养环境下大部分可存活。结论:壳聚糖温敏水凝胶具有良好的生物相容性,有望应用于牙周组织工程。Objective To evaluate the biocompatibility of thermosensitive chitosan hydrogel with periodontal ligament cells. Methods The periodontal ligament cells (PDL cells) were inoculated into the chitosan hydrogel scaffold for 3D culture. MTT, flow cytometry, cell viability staining was performed to evaluate the biocompatibility of the PDL cells cultured in the 3D hydrogel scaffold. Results By adding β-glycerophosphate (β-GP) into chitosan solution, gelation of thermosensitive hydrogel can be obtained under 37℃ within 30 min. Chitosan hydrogel extractions didn' t inhibit PDL cell proliferation, and could increase the percentage of S-phase cells. Cell viability staining demonstrated that most of the PDL cells could survive in the chitosan hydrogel scaffold. Conclusion The thermosensitive chitosan hydrogel has excellent biocompatibility, which may be a promising injectable material for periodontal ligament tissue engineering.
