摘  要:目的观察不同浓度的雌激素作用于体外培养的人胚髁突软骨细胞(MCCs),探讨雌激素对MCCs增殖分化的影响。方法体外培养MCCs,甲苯胺蓝及Ⅱ型胶原免疫细胞化学染色法鉴定;甲基噻唑基四唑(MTT)法检测不同浓度雌激素对MCCs增殖与分化的影响。结果高浓度(10-6mol·L-1)、低浓度(10-12mol·L-1)雌二醇抑制MCCs增殖分化,生理浓度(10-10、10-8mol·L-1)雌二醇促进MCCs增殖分化,且随作用时间延长,促进或抑制作用增强。结论雌激素对体外培养的人胚MCCs增殖分化具有双向调节作用,呈时间和剂量依赖性。表明雌激素对维持颞下颌关节的正常功能具有重要意义,可能在颞下颌关节病的发生和进程中发挥作用。Objective To determine the effects of different estrogen concentrations on the proliferation and differentiation of human embryo mandibular condylar chondrocytes (MCCs). Methods In vitro-cultured MCCs were incubated and treated with estrogen at different concentrations. The MCCs were identified by toluidine blue and type II collagen immunocytoche- mical methods, and cell proliferation was monitored using methyl thiazolyl tetrazolium assay. Results The addition of 10^-12 or 10^-6 mol·L^-1 exogenous estrogen inhibited the growth of MCCs, whereas addition of 10^-10 or 10^-8 mol·L^-1 exogenous estrogen promoted growth. These estrogen effects were time-dependent. Conclusion Estrogen can affect cellular MCCs proliferation and differentiation in a biphasic manner depending on the concentration. Estrogen may be vital in the maintenance of normal functions of the temporomandibular joint (TMJ) and may be involved in the progression of certain TMJ diseases.
