神经营养因子-3在成年大鼠胃和睾丸分布的研究

摘　　要：用特异的神经营养因子 - 3(NT- 3)兔抗血清 ,以免疫组化 ABC法染片观察了 NT- 3样免疫反应物(NT- 3- IR)在成年大鼠胃和睾丸的分布。结果 :NT- 3- IR主要分布于胃的部分上皮细胞、壁细胞、主细胞和平滑肌细胞。阳性产物的亚细胞定位除胞浆外 ,亦有细胞核内染色 ;NT- 3- IR在睾丸主要分布于曲精小管上皮的部分各级生精细胞 ,阳性染色位于细胞浆。结果提示 :NT- 3可能与胃和睾丸的某些生理功能有关 ,且两者发挥功能的方式可能有异。The bulk of evidences have demonstrated that Neurotrophin 3 (NT 3) and its mRNA were found in not only central nervous system but also peripheral nervous system. The effect of NT 3 in maintaining neuronal survival and promoting neurite growth has been well known. Moreover, some evidences on the distribution of NT 3 in non nervous system, such as intestine, kidney and spleen were provided. In this study, we investigated the distribution of NT 3 like immunoreactivity (NT 3 IR) in stomach and testis in adult rats using specific antiserum of NT 3 by immunochemistry ABC method. The results were as follows: The positive immmunoreactive products of NT 3 were observed in stomach epithelium cells, paracytes, chief cells, and smooth muscle cells. The subcellular localization for NT 3 IR is mainly in nucleus and cytoplasm. Comparatively, NT 3 IR in testis is mainly distributed in some spermatogenic cells including many spermatogoniums, primary spermatocytes, secondary spermatocytes and spermatids. The positive immunoreactive products are mainly located in cytoplasm. Specifically, NT 3 IR is also found in base membrane. These results suggested that there might be difference in the mechanism of the NT 3 between stomach and testis deduced from the diversity of subcellular locations for NT 3 IR.
