摘 要:目的:观察荞麦花叶总黄酮对压力超负荷性大鼠心肌肥厚的影响及其可能机制。方法:腹主动脉缩窄法建立大鼠压力超负荷心肌肥厚模型,设假手术组、模型组、卡托普利组(10mg/kg)及荞麦花叶总黄酮低、中、高剂量组(100、200、400mg/kg)。造模1周后灌胃给药,给药8周后测量大鼠心功能变化如左室收缩内压(LVSP)、左室舒张末期压(LVEDP)、左心室内压最大上升/下降速率(±dp/dtmax);计算心脏质量指数(HMI)以及左心室质量指数(LVMI);比色法测定超氧化物歧化酶(SOD)、丙二醛(MDA),ELISA法检测IL-6、IL-10及TNF-α。结果:与模型组比较,荞麦花叶总黄酮能明显降低心功能各项指标及HMI、LVMI;升高血清SOD、IL-10水平,降低MDA、IL-6、TNF-α水平。结论:荞麦花叶总黄酮具有抑制压力超负荷性大鼠心肌肥厚的作用,其机制可能与抗氧化和抗炎作用相关。To examine the effect of total flavones of buckwheat flower and leaf on myocardial hypertrophy induced by pressure overload in rats, as well as to explore the mechanisms involved. Methods: The model rats in which myocardial hypertrophy was induced with abdomi nal aortic banding. SD rats were randomly divided into 6 groups: Sham group,model group, eaptopfil group( 10 mg/kg), total flavones of buckwheat flower and leaf groups with various dosages ( 100, 200 and 400mg/kg) . After administration of the tested drugs for 8 weeks, LVSP, LVEDP and ± dp/dtmax were recorded. Heart mass index and left ventricular mass index were calculated. The content of SOD, MDA in serum were measured by colofimetric method. The level of IL-6,IL-10 and TNF-α in serum was examined by ELISA. Results: Com pared with model group, total flavones of buckwheat flower and leaf groups could significantly decrease indexes of cardiac function, HMI and LVMI, increase the levels of SOD and IL-10 in serum, and decrease the levels of MDA, IL-6, and TNF-α in serum. Conclusion: Total fla vones of buckwheat flower and leaf significantly attenuates myocardial hypertrophy induced by pressure overload in rats. The possible mecha nisms are in Dart associated with antioxwenation and anti-inflammatory action.
