摘  要:目的 探讨大鼠抑郁模型的行为学及海马磁共振T2信号强度比的动态变化及两者的相关性.方法 30只SD大鼠根据行为学评分后,随机分为对照组和抑郁组,每组15只.运用慢性不可预知应激及孤养模式建立抑郁模型,应激共8周.于建模开始前及每周末进行大鼠体质量测量、液体消耗及Morris水迷宫试验,并于实验开始前及每2周末在高场强MRI上测量每组大鼠各侧海马CA1-3区的T2信号变化,并计算与额叶的信号强度比.结果 各项行为学指标表明建模成功并呈动态变化.与对照组相比,左侧海马各区T2信号强度比于第4周差异有统计学意义(CA1:抑郁组1.11±0.10,对照组1.25±0.13,P〈0.01;CA2:抑郁组1.11±0.08,对照组1.17±0.07,P〈0.05;CA3:抑郁组1.12±0.08,对照组1.28±0.16;P〈0.01),其中CA3区于第2周就明显增高;而右侧海马在变化时间及增高幅度较左侧迟缓.同时发现行为学指标与海马信号比改变具有一定的相关性.结论 大鼠抑郁模型的双侧海马信号存在异常,且各区表现出信号变化的特异性并呈动态变化.Objective To investigate the dynamic change and correlation between the behavior and the ratio of hippocampal T2 signal intensity by MRI of rat depressive model. Methods Based on the evaluation of ethology, 30 SD rats were randomly and equally divided into two groups: the control group and the model group which was exposed to chronic unpredictable mild stress for 8 weeks. The body weight, fluid consumption test and Morris water maze test were measured at the beginning and per week while the T2 signal of bilateral hippocampal CA1-3 of all the rats were measured by high field MRI at the beginning and per two weeks, and the ratio of signal intensity with frontal lobe was calculated. Results Based on the behavioral data, the rat depressive model has been established successfully, changing dynarnicly. On 4th week, the ratio of T2 signal intensity of left hippocampus was significantly heightened while that of CA3 was done on 2nd week(P〈0.05). And the ratio of T2 signal intensity of right hippocampal was weaker than the left one of the changing time and the degree. There was correlation apparently between the behavior and the ratio of hippocampal T2 signal intensity. Conclusion The bilateral hippocampal signal of rat depressive model is characteristically abnormal and changes dynamically.
