壳聚糖多孔珠作为亲和吸附剂载体纯化人尿胰蛋白酶抑制剂

摘　　要：UTI粗品经壳聚糖多孔珠 胰蛋白酶亲和层析 ,得到比活为 135 0U/mg ,收率为 40 .2 %UTI组分。再经SephadexG 10 0凝胶过滤 ,发现存在四种分子形式 ,其相对分子质量分别为 6 70 0 0 ,43 0 0 0 ,2 3 0 0 0 ,16 0 0 0。其中 ,16 0 0 0的无UTI活性。Column packed with trypsin chitosan porous beads affinity adsorbents was employed for crude UTI chromatography. The experimental results showed that the specific activity of the UTI could reach 1 350U/mg and the yield was 40.2%. The results also revealed the affinity adsorbents possessed nice mechanical strength and storage stability.The characterization of the affinity column had no change after repeatedly using six times. After purification of affinity chromatography,four molecular forms were separated .Their molecular weights were about 67000,43000,25000 and 16000 respectively. Among these,the molecular form of 16 000 had no activity.
