摘  要:目的探讨三氧化二砷联合氟尿嘧啶(5-FU)对肝癌细胞凋亡相关因子Bcl-2和Bax表达的影响。方法体外培养人肝癌SMMC-7721细胞株,将细胞分为四组,即As2O3组、5-FU组、As2O3+5-FU组及对照组,采用免疫细胞化学法检测各组SMMC-7721细胞Bcl-2、Bax蛋白的表达。结果 As2O3组、5-FU组、As2O3+5-FU组细胞Bcl-2蛋白表达较对照组明显降低,Bax蛋白表达明显升高(P<0.01);联合用药组细胞Bcl-2蛋白表达较单用药组明显降低,Bax蛋白表达明显升高(P<0.01)。结论 As2O3联合5-FU可下调Bcl-2蛋白的表达,上调Bax蛋白的表达,从而诱导肝癌细胞凋亡。Objective To explore the effects of arsenic trioxide combined with 5 - FU on the expressions of apoptosis factors Bcl- 2 and Bax of liver cancer cell line. Methods Cultured human liver cancer cell line SMMC-7721 was divided into four groups in vitro: As203 group, 5- FU group, As203 plus 5- FU group and control group, the expressions of Bcl- 2 and Bax of these cells were examined by immunocytochemistry. Results The expression of Bcl - 2 was significantly lower in the As203 group, 5 - FU group and Asz 03 plus 5 - FU group than the control group, the expression of Bax was significantly increased(P〈 0.01). The expression of Bcl - 2 was significantly lower in the combinational drug group than the single drug group, the expression of Bax was significantly increased (P〈 0. 01). Conclusion Arsenic trioxide combined with 5 - FU can induce apoptosis of liver cancer cells by downregulating the expression of Bcl - 2 and upregulating the expression of Bax
