摘  要:目的探讨诱导大鼠原位膀胱癌动物模型早期应用雌激素干预,观察其膀胱的成瘤情况、肿瘤大小及病理分级有无影响。方法给切除卵巢的SD大鼠行N-甲基亚硝基脲(MNU)膀胱灌注,每次2mg/只,1次/2周,共4次,以制备大鼠原位膀胱癌动物模型。随机分成雌激素提前干预、雌激素非提前干预和仅灌注MNU三组相比较。结果应用雌激素干预的二组与仅灌注MNU组相比较,在成瘤率、肿瘤大小及方面差异均有统计学意义(P<0.01);雌激素提前干预和雌激素非提前干预二组间比较差异无统计学意义(P>0.05),但雌激素提前干预组肿瘤发生率更低。结论化学致癌剂N-甲基亚硝基脲(MNU)经膀胱灌注可成功建立原位膀胱癌动物模型,经雌激素提前干预明显降低膀胱癌的成瘤率及抑制肿瘤生长,为膀胱癌内分泌治疗提供佐证。Objective To discuss the impact of early estrogen intervention on tumor formation, tumor size, histological grade for animal model rat of orthotopic bladder cancer. Methods The ovariectomized SD rats were selected as experimental application. Experimental group underwent bladder perfusion with N - methyl nitrosourea {MNU) to prepare animal model of orthotopic bladder cancer, once every two weeks, every 2mg, a total of four times, each with MNU 8mg. The effects were compared in early estrogen intervention, estrogen non early intervention and the only perfused MNU groups. Results There were differences in tumor rate, tumor diameter and histological grade among estrogen therapy groups and the group of only bladder perfusion with MNU( P〈0. 01). Whether estrogen in advance between the two groups was not statistically significant( P〉0.05 ), but the incidence of cancer with early estrogen intervention was lower. Conclusions Intravesical bladder with a chemical carcinogen MNU can successfully establish orthotopic cancer animal model. The early estrogen intervention can significantly inhibit tumor growth, provide evidence of endocrine therapy for bladder cancer.
