摘 要:为研究有利于E3菌株产N6-(2-羟乙基)腺苷[N6-(2-hydroxyethyl)-adenosine,HEA]的培养基组分、前体物及氨基酸,以查氏培养基作为基础培养基,液体静置发酵,分别对碳氮源、无机盐、前体物和氨基酸进行筛选,结果表明:碳、氮源最好的是淀粉和(NH4)2HPO4;无机盐只有钾盐能促进菌株产生HEA,效果最好的是KH2PO4;前体物和氨基酸中腺苷和腺嘌呤促进菌株累积HEA能力相同,精氨酸、赖氨酸、脯氨酸、组氨酸、色氨酸、L-谷氨酸和丝氨酸等均能促进菌株产HEA,精氨酸效果最好,菌丝体中HEA含量达2068.21±55.34μg/g,HEA产量达13.10±1.48mg/L。N6-(2-hydroxyethyl)-adenosine (HEA) production by Isaria tenuipes, strain E3, grown on media containing different carbon and nitrogen sources, and supplemented with different amino acids, inorganic salts and chemical precursors, was evaluated. Highest HEA production was recorded on Czapek medium (without sucrose or NaNO3 ) when starch and (NH4)2 HPO4 served as carbon and nitrogen sources, respectively Potassium phosphate promoted HEA production when added to Czapek medium containing 3% sucrose and 0.2% (w/v) NaNO3. Supplementation of Czapek medium with adenosine, adenine, arginine, lysine, proline, histidine, tryptophan, L-glutamic acid or serine also increased HEA production. Adenosine and adenine increased HEA yields by ~380%, while HEA levels of 2068.21 +- 55.34 /~g/g were recorded in mycelia harvested from cultures supplemented with arginine (the most effective amino acid for enhancing HEA production by strain E3) compared with 148.56 -+ 4.78 g in mycelia from unsupplemented controls. Combined mycelium and culture fluid from one liter of arginine-supplemented culture contained up to 13.10 + 1.48 mg/L HEA, representing a 17.2-fold increase over controls.
