黄荆子乙酸乙酯提取物对人乳腺癌MCF-7细胞凋亡的影响

摘　　要：目的研究黄荆子乙酸乙酯提取物(EVn-50)诱导人乳腺癌MCF-7细胞凋亡作用。方法分3组:空白组(0.02%DMSO)、试验组(EVn-50)、阳性对照组(紫杉醇)。量效测定:试验组给予低、中、高浓度(0.3,3.0,30.0μg·mL-1)的EVn-50,阳性对照组给予紫杉醇(0.2,2.0,20.0μg·mL-1),培养48h。时效测定:2组均给予中浓度,分别培养24,48,72 h。AO/EB双染色荧光显微镜观察凋亡细胞形态;FITC标记Annexin-V/PI双染流式细胞术分析早期凋亡细胞百分率;DNA琼脂糖凝胶蛋白电泳观察MCF-7细胞DNA核小体间断裂的梯形条带。结果在3个时间点Evn-50组均可见细胞凋亡,与紫杉醇组的凋亡率分别为31.83%,47.83%(24 h);51.83%,76.33%(48 h);85.67%,98.5%(72 h)(均P<0.05),低、中、高浓度的Evn-50组与紫杉醇组的凋亡率分别为13%,23.33%(低);51.83%,76.33%(中);55.17%,99.17%(高)(均P<0.05)。DNA在48,72 h有典型梯形条带。结论 EVn-50有诱导人乳腺癌MCF-7细胞凋亡的作用。Objective To study the effects of the acetoacetate extract of Vitex negundo seeds （ EVn - 50） induced by human breast cancer MCF - 7 cell apoptosis. Methods There were three groups： blank group （ 0. 02% DMSO ）, the trial group （ EVn - 50 ） and control group （ paclitaxel）. Dose- effect determination： trial group were treated with low, middle and high concentration （0. 3,3.0 ,30. 0μg·mL^-1） of EVn - 50, the positive control group received paclitaxel （ 0. 2,2. 0 , 20. 0 μg·mL^-1）, cultured for 48 hours. Aging test ： the trial group and positive control group were both given in the medium concentration, cultured for 24, 48 and 72 hours. AO/EB double staining fluorescence microscopy to counting the typical apoptotic morphology of ceils in percentage. FITC - Annexin - V/PI double staining by flow cytometry to analyze the percentage of early apoptotic cells. DNA agarose gel eleetrophoresis MCF -7 cells, to observe DNA ladder changes. Results There be seen apoptosis in Evn - 50 group at 24, 48 and 72 hours. The apoptotic rates in Evn - 50 group and paclitaxel group were 31.83%, 47.83% （24 h）; 51.83%, 76.33% （48 h） ; 85.67% ,98.5% （72 h）, respectively, have statistically difference （ P 〈 0. 05 ）. At the low, middle and high concentration, the apoptotic rates on the same concentration in Evn -50 group and paclitaxel group were 13%, 23.33% （low） ; 51.83/76.33% （middle） ; 55.17%, 99. 17% （high） , respectively, have statistically difference （ P 〈 0. 05 ）. DNA at 48 and 72 hours have typical ladder. Conclusion EVn - 50 can induce human breast cancer MCF-7 cell apoptosis.
