摘　　要：目的探讨Sharpin蛋白在人不同前列腺癌细胞株中与前列腺癌组织中的表达及其与Gleason评分、血清PSA的关系。方法采用实时荧光定量PCR法,检测Sharpin在DU145、PC-3和LNCaP 3种常见的前列腺癌细胞株和RWPE-1正常前列腺上皮细胞株中的表达。同时采用免疫组织化学方法检测Sharpin在前列腺增生及前列腺癌组织中的表达,并探讨与临床病理特征的关系。结果 Sharpin在3种前列腺癌细胞株中的mRNA水平(1.62±0.31,1.36±0.23,2.1±0.1)要明显高于正常前列腺上皮细胞RWPE-1(0.6±0.11)。免疫组织化学结果示Sharpin在前列腺癌组织中高表达,前列腺癌中的阳性表达率远远高于前列腺增生组织,平均染色得分也要远远高于前列腺增生组织。另外,Sharpin在前列腺癌组织中的表达与患者的Gleason评分和术前血清的t-PSA密切相关,均呈正相关(P<0.05)。结论 Sharpin可能是前列腺癌的肿瘤相关抗原,sharpin的表达可能具有评估前列腺癌患者病情、指导临床治疗方案的指导及判断预后及复发的作用。Objective To explore the expression of Sharpin in different human prostate cancer cell lines and tissues and the relationship with Gleason Grade and serum t-PSA. Methods Quantitative RT-PCR and Western Blot were performed to detect the expression of Sharpin mRNA and protein in three common prostate cancer cell lines including DU145, LNCaP and PC-3 and normal prostate epithelial cell line RW-PE-1. Then we used immunohistochemistry to investigate the expression of Sharpin protein in tissues of prostate cancer and benign prostate hyperplasia, and the relationship with Gleason grade and serum PSA was analyzed. Results The level of Sharpin mRNA in three common prostate cancer cell lines were 1.62±0.31,1.36±0.23 and 2.1±0.1 respectively, higher than that of RWPE-1 cell line（0.6±0.11 ）. And the Western Blot showed that the expression of Sharpin in prostate cancer cell lines are much higher than normal prostate epithelial cell line. By means of immunohistoehemistry, we observed that Sharpin exists in most prostate cancer tissue because the rate of Sharpin positive expression in prostate cancer was much higher than benign prostate hyperplasia. In addition, we found that the level of Sharpin in prostate cancer was significant correlated with Gleason Grade and serum t-PSA （P〈0.05）. Conclusions Sharpin may be a novel tumor-associated antigen of prostate cancer. Sharpin may be a novel indicator for evaluating the severity, deciding the therapeutic regimen and prognosis and recurrence in prostate cancer.
