摘  要:目的研究胰岛素对血管性痴呆(VD)大鼠海马NMDA受体及突触可塑性的影响。方法采用结扎双侧颈总动脉法复制VD大鼠模型,将60只SD大鼠随机分为治疗组(20只)、模型组(20只)和假手术组(20只)。治疗组经侧脑室注射胰岛素(1 IU/kg),模型组、假手术组注入生理盐水5μL。术后28 d开始行水迷宫实验评估大鼠学习记忆能力;术后1 d及35 d用免疫组织化学法及免疫蛋白印迹法检测大鼠海马NMDA受体表达量;术后35 d用免疫蛋白印迹法检测大鼠海马突触素表达量。结果水迷宫实验提示治疗组学习记忆能力优于模型组;术后1 d,治疗组、模型组和假手术组大鼠海马NMDA受体灰度值分别为(125.44+8.35)、(110.28+13.67)和(145.12+17.23),差异有统计学意义(F=16.54,P<0.05),大鼠海马NMDA受体相对表达量治疗组为假手术组的1.48倍,模型组为假手术组的2.28倍(P<0.05);术后35 d,治疗组、模型组和假手术组大鼠海马NMDA受体灰度值分别为(155.63+13.48)、(170.79+11.13)和(139.12+18.25),差异有统计学意义(F=11.79,P<0.05),大鼠海马NMDA受体相对表达量治疗组为假手术组的0.72倍,模型组为假手术组的0.48倍(P<0.05);大鼠海马突触素相对表达量治疗组为假手术组的0.72倍,模型组为假手术组的0.46倍(P<0.05)。结论胰岛素可能通过调节VD大鼠海马NMDA受体及突触素的表达,改善VD大鼠的认知功能。[Objective] To investigate the effects of insulin on hippocampus NMDA receptor (NMDAR) and synaptic plasticity in the rats with vascular dementia. [Methods] 60 male Sprague-Dawley rats were randomly assigned to a treating group (n =20), a model group (n =20), and a sham-operation group (n =20). The rats in the former 2 groups were operated on to establish the experimental vascular dementia model by bilateral common carotid artery permanent ligation. Insulin was injected intracerebroventricularly in treating group, and saline was injected intracerebroventricularly in model and sham-operation group. The rats' learning and memory were assessed at 4 weeks after operation. Immunohistochemical staining was used to detect NMDAR expression in hippocampus. Western blotting was used to detect NMDAR and synaptophysin. [Results] The rats in treating group showed significantly better learning ability than model group (P 〈0.05). At the first day after operation, the grey level of NMDAR in the hippocampus of the rats for the model group is (110.28 ±13.67), (125.44 ±8.35) for the treating group and (145.12±17.23) for the sham-operation group, with obvious differences among the groups (F =16.54, P 〈0.05). The expression level of NMDAR of treating group is 1.48 times as sham-operation group (P 〈0.05), the expression level of NMDAR of model group is 2.28 times as sham-operation group (P〈0.05). At 35 days after operation, the grey level of NMDAR in the hippocampus of the rats for the model group is (170.79 ±11.13), (155.63 ±13.48) for the treating group and (139.12 ±18.25) for the sham-operation group, with obvious differences among the groups (F=11.79, P〈0.05).
