摘 要:目的研究Neu-p11预处理对大鼠心肌缺血/再灌注损伤细胞凋亡的影响。方法 30只♂SD大鼠随机分为3组:假手术组,缺血/再灌注损伤组和Neu-p11预处理组。测定血清中乳酸脱氢酶(LDH)、肌酸激酶(CK)的活性及心肌丙二醛(MDA)的含量,TUNEL法检测心肌细胞凋亡指数,免疫印迹法检测Bcl-2、Bax蛋白的表达。结果 Neu-p11预处理可明显改善缺血/再灌注损伤大鼠的心功能损害,减少血清中LDH、CK的释放和心肌组织中MDA的产生,可使心肌细胞凋亡率减少,心肌Bcl-2蛋白表达上调,Bax蛋白表达下调。结论 Neu-p11对心肌缺血/再灌注损伤具有保护作用,其机制可能与抑制细胞凋亡有关。Aim To study the effect of Neu-p11 on myocardial ischemia-reperfusion injury in rats. Meth- ods Thirty male Wistar rats were randomly divided into three groups: sham group,I/R group and Neu-p11 group. LDH and CK activity in serum and MDA con- tent in myocardial tissue were measured. Cell apoptosis was evaluated by TUNEL assay. Bcl-2 and Bax protein expressions were detected by Western blot. Results Neu-pl 1 significantly decreased the level of LDH, CK and MDA content. The myocardial apoptotic index in Neu-pll group was markedly lower than that in I/Rgroup. And Western blot analysis revealed that the ex- pression of Bcl-2 significantly increased, while Bax ex- pression significantly decreased in Neu-pll group, compared with those in I/R group. Conclusion Neu- pll shows protective effects against myocardial ische- mia-reperfusion injury in rats, and the mechanism is probably related to its anti-apoptotic activity.
