Effect of Swimming Exercise on Hypothalamus SOCS -3, BDNF and Leptin -resistance in Rat with High -fat Diet

摘  要:目的:观察运动对高脂饮食大鼠血清和下丘脑瘦素(Leptin)以及下丘脑瘦素长型受体(OB—Rh)、促黑皮素受体4(MC4R)、细胞因子信号转导抑制因子-3(SOCS-3)mRNA表达和脑源性神经营养因子(BDNF)的影响,探讨适宜运动对高脂饮食大鼠预防肥胖的部分中枢机制。方法:雄性sD鼠24只,随机分为正常对照组(C组),高脂模型组(H组)和高脂60min运动组(HE组),每组8只。高脂模型组和高脂60rain运动组大鼠喂饲高脂饲料,高脂饮食运动组大鼠进行60min的无负重游泳运动。持续10周。结果:10周运动后,高脂模型组大鼠较正常对照组大鼠体重、Lees指数、体脂、脂体比均显著增高(P〈0.05),血清瘦素水平显著升高(P〈0.05),下丘脑内OB—Rb、MCARmRNA表达量显著降低(P〈0.05),SOCS-3mRNA表达量显著升高(P〈0.05),下丘脑BDNF含量显著降低(P〈0.05);高脂60rain运动组较高脂模型组大鼠体重显著降低(P〈0.01),LeeS指数、体脂、脂体比均显著降低(P〈0.05),血清瘦素水平显著升高(P〈0.05),下丘脑OB—Rb、MC4R、leptinmRNA表达量显著升高(P〈0.05),下丘脑SOCS-3mRNA表达量显著降低(P〈0.05),下丘脑BDNF含量显著升高(P〈0.05)。结论:长期适宜的运动可通过降低下丘脑SOCS-3的表达,增加下丘脑Leptin与受体OB—Rb的结合,降低了瘦素抵抗,激活下丘脑含MC4R受体的神经元,从而刺激BDNF的释放,从而起到预防高脂饮食引起肥胖的作用。Objective : To explore the possible mechanism of exercises in high - fat diet induced rats with obesity, we observed the serum leptin, the mRNA expression of leptin, OB - Rb, MC4R and SOCS - 3 in hypothalamus, and the content of BDNF in hypothalamus. Method: 24 male Spragne -Dawley rats were randomly assighned to 3groups (each n = 8 ) : control group (C Group), high - fat model group (HE) and high - fat 60 - min exercise training group ( HE Group), respectively. Rats of HE Group swam for 60 min each day without loading. Results : After 10 weeks, to compared with the C group, the body weigh, Leeg index and the body fat of H group were obviously increased (P 〈 0. 05 ) , the serum leptin was significantly increased (P 〈 0.05 ) ; In hypothalamus, the mRNA expression of OB - Rb and MCAR were dramatically decreased ( P 〈 0.05 ) and the mRNA expression of SOCS - 3 was markedly increased ( P 〈 0. 05 ) , the concentration of BDNF was obviously decreased ( P 〈0.05 ). To compared with the H group, the body weight, Lee~ index and the body fat of HE group was obviously lower ( P 〈 0.01 ) ; the serum leptin was significantly decreased ( P 〈 0.05) ; In hypothalamus, the mRNA expression of OB - Rb, leptin, and MCAR were dramatically increased ( P 〈 0.05 ), and the mRNA expression of SOCS - 3 markedly decreased ( P 〈 0.05 ), the concentration of BDNF was obviously increased ( P 〈 0.05 ). Conclusion : The appropriate long - term exercise can amelio- rate the "leptin resistance", the possible mechanisms is : a long - term exercise can decrease the expression mRNA of SOCS - 3, then induced OB -Rb binding to leptin, with its activation to the hypothalamic neurons MC4R that causing a increase in BDNF production, thereby reducing food intake and stimulating the motor neuron to increase exercise training, so that in- creased energy expenditure.
