摘 要:目的探讨细胞周期蛋白Cullin-1(Cul-1)和S期激酶相关蛋白2(Skp2)在鼻咽癌及癌前病变组织中的表达及意义,同时对比它们在鼻咽癌及癌前病变组织中的表达水平,比较二者的分布情况,找出其差异性及相关关系。方法应用免疫组化方法检测Cul-1和Skp2蛋白在20例分化型癌、20例未分化癌和20例鼻咽癌前病变中的表达。结果在20例分化型癌、20例未分化癌和20例鼻咽癌前病变中Cul-1的平均阳性表达率分别为76%、77%、54%;阳性表达值分别为1.63±0.7、1.62±0.68、1.15±0.64,Cul-1在2种鼻咽癌中的表达高于癌前病变,差异有统计学意义(P<0.05),2种鼻咽癌之间无差异;Skp2在20例分化型癌、20例未分化癌和20例鼻咽癌前病变中的平均阳性表达率为32%、41%、33%,平均阳性表达值分别为0.53±0.49、0.82±0.54、0.72±0.43,无统计学差异。对20例分化型鼻咽癌标本Cul-1、Skp2阳性表达值进行相关关系分析,两者相关性不显著。结论 Cul-1参与了鼻咽癌发生、发展,Skp2则可能参与了鼻咽癌的发生过程,它参与的细胞周期调节可能是鼻咽癌发生的早期事件;两种蛋白因子在细胞周期调控中的关系有待进一步验证。Objective To detect the expression of Cullin-1 (Cul-1) and S-phase kinase associated protein 2 (Skp2) in the tissues of nasopharyngeal carcinoma (NPC) and precancerous lesion, and elucidate the relationship between them. Methods Immunohistoehemical methods were taken to detect the expression of Cul-1 and Skp2 in the tissues taken before treatment among 20 differentiated carcinoma patients, 20 undifferentiated carcinoma patients, and 20 precancerous lesion patients. At last, the relationship between the expression of these protein was analyzed. Results The positive expression rates of Cul-1 in the tissues of differentiated carcinoma, undifferentiated carcinoma and precancerous lesion were 76%, 77% and 54%, respectively. The expression value were 1.63±0.7, 1.62±0.68, 1.15±0.64, respectively. The differences between differentiated carcinoma and precancerous lesion, undifferentiated carcinoma and precancerous lesion were statistically significant (P〈0.05). The positive expression rates of Skp2 in the tissues of differentiated carcinoma, undifferentiated carcinoma and precancerous lesion were 32%, 41%, 33%, respectively. The expression value were 0.53±0.49, 0.82±0.54, 0.72±0.43, and there were no difference between them. To analyze the expression of Cul-1 and Skp2 in 20 differentiated carcinoma, which had no significant correlation. Conclusion Cul-1 and Skp2 protein were involved in the occurrence and development of human nasopharyngeal carcinoma. And the Skp2 participated in cell cycle regulation that may be an early event of nasopharyngeal carcinoma. The relationship between Cul-1 and Skp2 in cell cycle regulation need further validation.
