异构性固化瘤苗(溶癌因子)研究与临床免疫治疗肺癌的观察

摘　　要：目的　研究肿瘤的特异性细胞免疫。方法　应用分子生物学和基因工程技术 ,由同种异体同源癌细胞中提取全息性、异构性固化瘤苗 (Heteroiditysolidifiedtumorvaccine HSTV) ,进行动物 (小鼠 )细胞免疫功能测试和应用肺腺癌 (LA795 )制备HSTV免疫小鼠 (T739)抗肺癌 (LA795 ) 3× 10 8个癌细胞与对照组进行比较及临床用肺癌SHTV免疫治疗肺癌 5 3例 (志愿者 )对其疗效进行观察。结果　发现具有类PHA和类LI 2作用的HSTV。免疫小鼠抗肺癌 (LA795 )有效率 72 %。临床中晚期肺癌术后应用HSTV ,一年抗复发率 86 .4%(对照组 5 7.2 %)。三年抗复发率 72 %,比对照组提高 3倍 ,且生活质量明显提高。对晚期患者应用HSTV ,能延长生存期和抗癌性疼痛。认为HSTV属一种全息性抗原肽 ,它通过在机体内三分子复体免疫机制 ,可实现激活癌症患者产生 30 0～ 5 0 0倍自主特异性细胞免疫的临床抗癌效应。Objective To study the specific cellular immunity of tumors.Methods Holographic heteroideous solidified tumor vaccine (HSTV) was extracted from homologous cancer cells with molecular biology and gene engineering techniques.The immune action of HSTV was examined in T739 mice.3×10 8 LA795 human lung cancer cells were injected into SHTV immunized T739 mices and the findings were compared with those of the control.53 volunteers of patients with lung cancer in advanced and middle stages were treated with HSTV.Results It was found that HSTV possessed actions similar to those of PHA and interleukin 2.72% of T739 mice immunized with HSTV were resistant to the injection of LA795 lung cancer cells.In the 53 patients with advanced lung cancer,the one year survival rate after treated with HSTV was 86.4%(57.2% in the control)and the threee year survival rate was 72%(3 times higher than that of the control).In addition,HSTV improved life quality,prolong survival term and alleviated the pain due to carcinoma.Conclusions HSTV, a kind of holoinformative antigen peptide, can activate autospecific cellular immunity 300～500 times higher and consequently enhance clinical anticancer effects through the immune mechanism of 3 molecule complex.
