摘  要:目的:建立可重复性大鼠脑胶质瘤模型。方法:雄性SD大鼠30只,体重150~250克,分为脑胶质瘤组(20只)和对照组(10只)。于前囟前1mm、矢状缝右旁开3mm处用牙钻钻一骨孔,用静脉注射套管针立体定向注射10ul含有约10~6个C6细胞的混悬液于鼠脑右侧尾状核内,建立鼠脑胶质瘤模型。对照组手术过程同上,但只注射10ul细胞培养基。两组大鼠于术后第16、25天分别作MRI测量胶质瘤大小、位置。术后第25天处死大鼠,立刻取脑,制作病理切片,观察组织形态学变化。脑胶质瘤组的石蜡切片经免疫组织化学染色检测S-100蛋白。结果:脑胶质瘤组,20只大鼠经MRI平扫、增强扫描和形态学检查证实均有单灶肿块,且肿瘤部位、大小、形态基本一致,无脑外转移和颅外扩散,经免疫组织化学染色检测S-100蛋白均为阳性。对照组均无瘤体。结论:用静脉注射套管针定向植入C6细胞到大鼠右侧尾状核,能产生可复制的单灶肿块,且无脑外转移和颅外扩散。0.5T MRI平扫和增强扫描能动态观察大鼠脑胶质瘤的生长情况。Objective:To establish a reproducible rat brain-glioma model. Methods:We divided 30 male SD rats,each weighing 150-250g,Separated into two groups, the experimental group (20 rats) and the control group (10rats). In experimental group, a bone hole was made by using a diamond-tipped burr at the following coordinates: anterior 1mm from bregma and lateral 3mm relative to the midsagittal sinus, a 10ul solution containing 106 C6 gliomaous cells was stereotactically implanted into right caudate nuclei to establish brain glioma model. The implanted process of control group was as same as the experimental group, but only a 10ul cell medium was injected. MRI was performed at the 16th and 25th day after implantation. The rats were sacrificed at the 25th day after implantation and the brain was extracted. Morphology and microscopic structure were observed in HE stain sections and S-100 protein was found in immunohistochemistry stain sections. Results:In experimental group, all 20 rats have unifocal masses and S-100 protein without extracerebral metabasis and the extracranial extension. The size, the site and morphology of all tumors are almost the same. No mass was found in all 10 rats of the control group. Conclusion:C6 gliomaous cells can be stereotactically implanted into the right caudate nuclei of the rats which may reproduce unifocal masses without extracerebral metabasis and extracranial extension. Gadolinium-enhanced MRI scan can follow the growth of the rat brain glioma.
