摘 要:【目的】研究辽东丁香枝体外抗氧化能力及对小鼠酒精性肝损伤的保护作用,为该植物的开发利用提供参考。【方法】制备辽东丁香枝乙醇提取物,通过对其总酚含量以及清除DPPH、ABTS+·能力和还原性能的测定,评估辽东丁香枝醇提取物的体外抗氧化活性;建立小鼠酒精性肝损伤模型,灌胃高(20g/kg)、中(10g/kg)、低(5g/kg)剂量(以生药计)的辽东丁香枝水提取物,研究其对小鼠肝脏系数、血清ALT和AST活性及肝组织中MDA含量的影响。【结果】辽东丁香枝醇提取物具有一定的抗氧化活性,其清除DPPH、ABTS+·的IC50值分别为0.070 1和0.308mg/mL;辽东丁香枝醇提取物具有较强的还原性,其总酚含量为64.8μg/mg。高、中、低剂量的辽东丁香枝水提物均能显著降低酒精性肝损伤小鼠血清中的ALT和AST活性以及肝组织中的MDA含量(P<0.05),其中高剂量灌胃还可使小鼠肝脏系数明显降低(P<0.05)。【结论】辽东丁香枝具有较好的体外抗氧化活性和保肝作用,并且对小鼠酒精性肝损伤保护作用的剂量依赖性不强,可将其开发为保肝新药。【Objective】This study aimed to study vitro antioxidant activities of Syringa wolfii C.K.Schneid branch and its protective effect on chronic alcoholic hepatic injury in mice,in order to provide reference for the development and utilization of this plant.【Method】Vitro antioxidant activities of alcohol extracts from S.wolfii branch were evaluated using DPPH and ABTS+· radical scavenging assays,and reducing power and total phenols content were determined.The ethanolinduced hepatocytes model was established to study the effects of gavaging high (20 g/kg),middle (10 g/kg),and low(5 g/kg) dosages (calculated by crude drug) of water extracts from S.wolfii branch on liver coefficient,ALT and AST activities in serum and MDA content in liver tissue of mice.【Result】The alcohol extracts from S.wolfii branch had comparable antioxidant activities,and the IC50 values for DPPH and ABTS+· radical scavenging assays were 0.070 1 mg/mL and 0.308 mg/mL,respectively.The alcohol extracts from S.wolfii branch exhibited strong reducing power, and the content of total phenols was 64.8 μg/mg. Serum ALT,AST activities,and MDA content in liver tissue of mice with alcoholic liver damage were significantly reduced by all dosage groups of water extracts from S.wolfii branch (P〈0.05).High dose remarkably decreased the liver coefficient (P〈0.05).【Conclusion】The branch of S.wolfii showed good vitro antioxidant activities and hepatoprotective effect,and the dependence on dose was not strong.Thus,it is a potential hepatoprotective new drug.
