摘 要:目的:比较非埋植型与埋植型种植体在完成义齿修复受载后种植体-骨界面中组织蛋白酶D(Cath-D)表达变化的异同。方法:Beagle犬8只,于下颌骨两侧分别植入非埋植型与埋植型种植体各3枚,第1枚种植体不做冠修复的种植体为对照,其余采用钴-铬合金铸造金属全冠修复。加载后2、4、8周分别处死动物,取材,采用免疫组化染色方法,观察种植义齿修复后不同时间点之间非埋植型与埋植型种植体骨界面Cath-D表达的灰度值动态变化。结果:2个对照组Cath-D表达不随时间变化(P>0.05),组间差异无显著性(P>0.05)。种植义齿修复受载后,2周时,种植体-骨界面Cath-D灰度值开始升高,但与对照组无差异;4周时达高峰,8周时表达略有降低,但4周和8周组均明显高于对照组(P<0.01);12周时恢复到接近对照组水平。所有非埋植型和埋植型种植体之间差异无显著性。结论:非埋植型和埋植型种植体在种植义齿修复受载后,引起种植体-骨界面Cath-D表达的变化,促进其界面的改建。Objective:To study the expression of cathepsin D (Cath-D) during implant-bone interface remodeling after loading of nonsubmerged or submerged implant dentures.Methods:Animal model was built in 8 adult beagle dogs.3 nonsubmerged and 3 submerged implants were respectively implanted into bilateral mandibles of the dogs.On each side 2 implants were restored with fixed metal full crowns to carry out the nonsubmerged or submerged implant dentures,1 implant without denture restoration was used as the control.The dogs were sacrificed 2,4 and 8 weeks respectively after loading.Cath-D expression in the implant-bone interface was examined by immunohistochemistry ABC method.Results:In the 2 control groups Cath-D expression was not changed with time(P >0.05) and there was no difference between the 2 control groups(P > 0.05).2 weeks after loading of implant dentures,Cath-D expression in the interface increased.4 weeks after loading,Cath-D reached the peak value vs control(P <0.05).8 weeks after loading Cath-D expression was still higher than that in the control(P < 0.05).It came down to the level of the control group 12 weeks after loading.There was no obvious difference of Cath-D expression in the implant-bone interface between the nonsubmerged and submerged implant dentures.Conclusion:Mechanical stress transferred from implant to surrounding bone tissue after loading of the nonsubmerged and submerged implant denture can stimulate Cath-D expression in the implant-bone interface and promote the interface remodeling.
