4NQO诱导大鼠口腔黏膜癌变过程中Bcl-2、BaxmRNA和蛋白的表达

摘　　要：目的：探讨Bcl-2、Bax mRNA和蛋白在4-硝基喹啉-1-氧化物（4-nitro-quinoline-1-oxide,4NQO）诱导大鼠口腔黏膜癌变过程中的表达及意义.方法：0.5％ 4NQO涂抹SD大鼠腭部黏膜,每周3次,持续18 ～ 26周,诱导大鼠口腔黏膜癌变,分别用逆转录-聚合酶链反应（RT-PCR）和免疫组化SABC法检测50例大鼠口腔黏膜癌变过程中Bcl-2、Bax mRNA和蛋白的表达.结果：Bcl-2 mRNA在正常（n=16）、异常增生（n=13）、鳞癌组织（n=21）中的相对值分别为0.31 ±0.02、0.56±0.09、1.03 ±0.13（P ＜0.05）,Bcl-2蛋白表达阳性例数分别是1/16、6/13、16/21 （P ＜0.05）.BaxmRNA在正常、异常增生、鳞癌组织中的相对值分别为0.62 ±0.05、0.68 ±0.04、0.28±0.14（P ＜0.05）,Bax蛋白表达阳性例数分别是5/16、4/13、3/21（P＜0.05）.结论：Bcl-2、Bax mRNA和蛋白异常表达与大鼠口腔黏膜癌变相关.Objective：To investigate the mRNA and protein expression of Bcl-2 and Bax in rat oral mucosal tissue during 4NQO-induced carcinogenesis.Methods：The rat oral mucosal carcinogenesis was induced by local infriction of 4NQO at 0.5％ three times a week for 18-26 weeks,the mRNA levels and protein expression of Bcl-2 and Bax in 50 rats mucoal tissue were detected by RT-PCR and immunohistochemical SABC method respectively.Results：The relative values of Bcl-2 mRNA in normal mucosa（n =16）,abnormal dysplasia（n =13） and oral squamous cell carcinoma（n =21） were 0.31 ± 0.02,0.56 ± 0.09 and 1.03 0.13 （P ＜ 0.05）,the protein expression of Bcl-2 were 1/16,6/13 and 16/21 （P ＜ 0.05）,respectively ; the relative values of Bax mRNA were 0.62 ± 0.05,0.68 ± 0.04 and 0.28 ± 0.14（P ＜ 0.05）,the protein expression of Bax were 5/16,4/13 and 3/21 （P ＜ 0.05）,respectively.Conclusion：Abnormal expression of Bcl-2 and Bax is involved in oral mucous carcinogenesis in rats.
