摘 要:目的:探讨吸烟对口腔黏膜细胞分泌β-防御素(hBD)的影响。方法:采用商业化可购的直接烟暴露装置,构建体外leuk1细胞吸烟模型,细胞吸烟时处于气液交换环境,将其分为吸烟组(吸入香烟烟雾5、10、20、40 min)和对照组(吸入含5%CO2空气10 min),采用酶联免疫吸附法检测hBD1、hBD2、hBD3的浓度。结果:hBD1在吸烟组表达明显低于对照组(P=0.004),hBD2在吸烟组表达高于对照组(P=0.012),而2组中hBD3分泌量差异无显著性。结论:吸烟可使口腔黏膜细胞分泌hBD发生改变,进而影响口腔黏膜免疫。Objective:To investigate the potential effects of cigarette smoke on the secretion of human beta-defensins(hBD) by oral mucosal cells.Methods:A commercially available smoking chamber was used to culture leuk1 cells at the air-liquid interface.Cells were divided into smoking group (exposed to cigarette smoke for 5,10,20 and 40 min respectively) and control group (exposed to 5% CO2 for 10 min).The level of hBD was detected by ELISA.Results:hBD1 in smoking group was significantly lower than in control group (P =0.004),however hBD2 expression in smoking group increased(vs control,P =0.012),and in the two groups the level of hBD3 had no statistical difference.Conclusion:Smoking may change hBD secretion by oral mucosal cells,and then influence the defense immunity of oral mucosa.
