摘 要:目的:研究高频重复经颅磁刺激(rTMS)对脑缺血大鼠缺血侧海马BDNF、VEGF和Nestin表达的影响。方法:SD雄性成年大鼠16只,随机分为模型组、rTMS组、rTMS+生理盐水(NS)组和rTMS+H89组各4只,rTMS+NS组和rTMS+H89组分别侧脑室注射NS和H89,4组均建立大脑中动脉栓塞(MCAO)模型,除模型组外均给予7 d 20 Hz rTMS治疗,应用Western Blot方法检测缺血侧海马pCREB、BDNF、VEGF和Nestin表达。结果:rTMS组pCREB、BDNF、VEGF和Nestin表达较模型组均增加(P<0.05),rTMS+H89组pCREB、BDNF、VEGF和Nestin表达较rTMS+NS组均减少(P<0.05)。结论:高频rTMS能使脑缺血后海马区BDNF、VEGF表达增加,并进一步促进Nestin表达,PKA-CREB通路的调节可能是其机制之一。Objective: To investigate the effects of high-frequency repetitive transcranial magnetic stimulation (rTMS) on the expression of BDNF, VEGF and Nestin in the ischemic hippocarnpus of rats with cerebral is- chemia. Methods: A total of 16 SD rats were randomly divided into model group, rTMS group, rTMS +H89 group and rTMS+normal saline(NS) group (n=4 in each group). The rats in rTMS+H89 group and rTMS+NS group were intracerebroventricular injected with H89 or NS respectively. Reperfusion models with middle cere- bral artery occlusion were established in the 4 groups, rTMS of 20 Hz was given to 3 groups except model group for 7 days. The expression changes of BDNF, VEGF and Nestin were investigated between model group and rTMS group, rTMS +H89 group and rTMS +NS group by Western Blot respectively. Results: Compared with model control group, the expressions of pCREB, BDNF, VEGF and Nestin were increased in the rTMS group (P〈0.05). Compared with rTMS+NS group, the expressions ofpCREB, BDNF, VEGF and Nestin were signifi- cantly decreased in the rTMS + H89 group (P〈 0.05). Conclusion: Repetitive transcranial magnetic stimulation of 20Hz could promote BDNF and VEGF expressions in the ischemic hippocampus after cerebral ischemia, which may contribute to the neurogenesis.
