摘 要:目的:探讨非小细胞肺癌(non-small cell lung cancer,NSCLC)组织Med19蛋白表达与DNA倍体和S期比例(S phase fraction,SPF)的关系。方法:收集山东省肿瘤医院2010-02-01-2011-07-31手术治疗、临床病理资料完整的56例NSCLC组织,应用免疫组化技术检测56例NSCLC组织中Med19蛋白水平,应用流式细胞术检测癌组织DNA倍体和SPF。结果:56例NSCLC组织中Med19阳性45例,阴性11例,二倍体14例,异倍体42例。14例二倍体肿瘤中Med19蛋白表达阴性6例,阳性8例;42例异倍体肿瘤中Med19蛋白表达阳性37例,阴性5例,异倍体肿瘤Med19蛋白阳性表达显著高于二倍体肿瘤,χ2=6.373,P=0.012。27例低SPF的患者中Med19蛋白表达阴性9例,阳性18例;29例高SPF的患者中Med19蛋白表达阴性2例,阳性27例,高SPF的患者肿瘤组织Med19蛋白阳性表达显著高于低SPF患者,P=0.018。结论:Med19作为潜在癌基因在NSCLC细胞增殖和多倍体形成中起一定作用,有望成为一个新的肿瘤治疗靶点。OBJECTIVE:To explore the relationship among MED19 protein expression,DNA ploid and SPF in non- small cell lung cancer(NSCLC) patient. METHODS: Fifyt cases of NSCLC in Shandong Cancer Hospital from Feb. 1,2010 to Jul. 31,2011 ,were involved in this study. By applying imunohistochemistry MED19 protein levels of 56 cancer tissues of NSCLC patients were detected. DNA ploidy of cancer tissue ~nd S-phase proportion(SPF) were detected by flow cy- tometry. RESULTS: In 56 cases, MED19 explession in 45 cases Were positive, 11 cases negative, 14 cases diploid and 42' ca- ses heteroploid. In 14 cases of diploid,8 cases were positive MED19 protein expression,6 cases were negative. In 42 cases of heteroploid, 37 cases were positive MED19 protein expression,5 cases were negative. The.positive MED19 protein ex- pression rate of heteroploid was markedly higher than that of diploid (X2=6. 373 ,P=0. 012). In 27 cases of low SPF pa- tient 18 cases were positive MED19 protein expression and 9 cases were negative. In 29 cases of high SPF patient 27 cases were positive MED19 protein expression and 2 cases were negative. The positive MED19 protein expression rate of high SPF was markedly higher than that of low SPF (P=0. 018). CONCLUSION: MED19 meets the definition of proto-onco- gene. As a potential oncogene it plays a role in the progression of lung cancer,and is expected to become a novel therapeu- tic target.
