摘 要:以环氧和氨基化的大孔树脂为载体,戊二醛为交联剂对木瓜蛋白酶进行了固定化,研究了不同载体和固定化条件对酶固定化率和相对活性的影响,考察了不同温度和pH值下游离酶和固定化酶的稳定性。实验结果表明,当给酶量为40mg,固定化温度为77℃,固定化时间为12h,pH值为6.5时,固定化酶的活性最高。与游离酶相比,固定化酶有更好的温度和pH值稳定性。Take the macroporous resin as carrier,the papain enzyme was immobilized using the glutaraldehyde as crosslinking agent,the influence of various carrier and preparation conditions on couple yield and relative activity of immobilized enzyme was studied,and the effect of temperature and pH on free and immobilized enzyme stability was also examined.As a result,the maximum relative activity of immobilized enzyme was obtained when enzyme amount was 40 mg,immobilized temperature was 77 ℃,immobilized time was 4 h and pH=6.5.The experiment result showed that the immobilized papain had better temperature and pH stability than free papain.
