摘 要:目的探讨JC病毒(JCV)早期基因编码产物大T抗原(large tumor antigen,T-Ag)的表达及β-catenin(β-连接素)蛋白表达与结直肠腺癌发生发展的关系。方法应用免疫组化法检测65例结直肠腺癌和20例非肿瘤胃黏膜中JCV T-Ag、β-catenin蛋白的表达。结果 65例结直肠腺癌中JCV T-Ag阳性表达率为38.4%(25/65)。20例非肿瘤结直肠黏膜中无JCV T-Ag蛋白阳性表达。结直肠腺癌中JCV T-Ag阳性表达与分化程度、浸润深度无统计学意义(P>0.05)。与淋巴结转移间有统计学意义(P<0.05),β-catenin蛋白在65例结直肠腺癌中正常表达率为47.7%(31/65),65例结直肠腺癌中弱阳性表达或不表达为52.3%(34/65)。结直肠腺癌中β-catenin蛋白异常表达与分化程度、浸润深度、淋巴结转移之间差异具有统计学意义(P<0.05),在20例非肿瘤胃黏膜100%阳性表达,即JCV T-Ag与β-catenin蛋白在结直肠腺癌组织中的表达呈负相关(P<0.01)。结论 JCV T-Ag早期基因编码产物大T抗原与β-catenin蛋白在结直肠腺癌的发生、发展中起重要作用。Objective To explore the role of JC virus (JCV) early region gene encoding product large tumor an- tigen( T-Ag ) and β-eatenin proteins expressions in colorectal adenocareinoma occurrence and development. Meth- ods The expressions of JCV T-Ag and β-eatenin proteins were detected by immunohistochemistry in 20 cases of nor- mal colorectal mucosa and 65 cases of colorectal cancer tissues. Results The positive rate of JCV T-Ag of colorectal adenocarcinoma was 38.4% ( 25/65 ) in 65 cases , and none of the JCV T-Ag was detected in the normal colorectal mucosa in 20cases. The positive expression of JCV T-Ag in the colorectal adenocarcinoma has no direct relationship to the differentiation or depth of infiltration ( P 〉 0. 05 ). But it was colsely correlated with lymph node metastasis ( P 〈0. 05 ). The positive rate of β-catenin of colorectal adenocarcinoma was 47.7% (31/65). The Weakly positive and negative rate( Abnormal expression) was 52. 3% ( 34/65 ) in 65 eases. The Abnormal expression of theβ-cate- nin was closely correlated with lymph node metastasis and differentiation or depth of infiltration. The normal expres- sion of β-catenin was detected in all normal colorectal mucosa in 20 cases. Conclusion JCV T-Ag early region gene encoding product large tumor antigen and β-catenin proteins were over-expressed in colorectal adenocarcinoma and their expressions had relevance to the carcinogenesis and the, progression of colorectal adenocarcinoma.
