摘  要:目的观察经口摄入全氟辛酸(PFOA)对大鼠血液抗氧化能力指标的影响,为分析PFOA的毒理学作用提供依据。方法将30只雄性SD大鼠随机分为3个剂量组(0、5、100 mg/kg BW),10只/组,PFOA经口染毒7 d,收集血液,测定血清总抗氧化能力、超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量及总谷胱甘肽(GSH)含量。结果PFOA染毒(100 mg/kg BW)导致大鼠体重降低、肝脏肿大、血清总抗氧化能力降低、脂质氧化产物MDA含量升高(P<0.05),但GSH水平变化差异无统计学意义(P>0.05),SOD活性则升高(P<0.05)。结论经口摄入PFOA对SD大鼠具有毒性,可导致肝脏损害,总抗氧化能力下降,但PFOA导致机体氧化损伤的作用机制尚需进一步研究证实。Objective To observe the effect of orally intake of perfiuorocaprylic acid (PFOA) on serum antioxidant capacity in SD rats. Methods Thirty male SD rats were randomly divided into three groups (0, 5 and 100 mg/kg BW, respectively) and orally exposed to PFOA for 7 days. Serum were collected and the serum total antioxidant capacity, serum superoxide dismutase (SOD) activity, malondialdehyde (MDA) and total glutathione (GSH) contents were determined. Results Exposure to PFOA for 7 days caused a significant decrease of body weight and liver enlargement. In 100 mg/kg BW group, the serum total antioxidant capacity was significantly decreased (P 〈 0.05 ) , lipid peroxidation product MDA concentration was significantly increased, but GSH level was only slightly decreased (P 〉 0. 05). Meanwhile, serum SOD activity was significantly increased after PFOA exposure. Conclusion PFOA is toxic to rats, leading to weight loss, liver damage and a decreased antioxidant capacity in male rats. However, the underlying mechanisms of oxidative damage caused by PFOA exposure still needs further elucidation.
