三刺皂荚黄化病植原体的分子鉴定

摘  要:采用巢式PCR,分别用植原体16S rRNA,16S-23S rRNA间区序列及tuf基因的通用引物对表现黄化症状的三刺皂荚DNA进行扩增,得到大小为1.2,0.3,0.8 kb特异性片段。对该片段分别克隆、测序及序列分析表明,三刺皂荚黄化病植原体(HoY)上述3序列与榆树黄化组B亚组(16SrV-B)的枣疯病植原体(JWB))相应序列的同源性最高,分别为99.8%,100%和100%。iPhyClassifier在线分析显示,HoY与JWB(AB052876)有相同的16S rDNA酶切图谱,表明三刺皂荚黄化病植原体属于榆树黄化组B亚组(16SrV-B)。Using nested PCR with universal primers for phytoplasmal 16S rRNA, 16S-23S rRNA space region and tuf genes, DNA fragments about 1.2 kb,0.3 kb and 0. 8 kb in length were amplified from the total DNA of honeylocust showing yellows symptom. Amplicons were cloned, sequenced and phylo- genetically analyzed. The results showed that the 16S rRNA, 16S-23S rRNA space region and tuf gene of phytoplasma isolated from honeylocust yellow phytoplasma (HoY)shared 99.8%, 100% and 100% identities with that of jujube witches'-broom phytoplasma (JWB) belonging to subgroup B of 'elm yellow group' (16SrV-B). The result of online analysis of iPhyClassifier for 16S rDNA showed the 16S rRNA gene of HoY had identical patterns with JWB (AB052876). All these results indicated that the phytoplsama associated with honeylocust yellows disease belonged to the subgroup B of 'elm yellows grouo' (16SrV-B).
