摘  要:川桑(M.notabilis Schneid.)是分布在四川、云南等山区的珍稀濒危单种桑树,资源稀少,单倍体,是桑树研究的重要材料.实验的目的是通过植物组织培养手段培养川桑离体再生植株,保存川桑种质资源及展开桑树的基础研究.实验内容包括了选择外植体、优化培养基成分和探讨生长调节物质对川桑离体再生的影响.结果表明:茎段容易被污染、叶片只能诱导出愈伤组织、以腋芽作为外植体大大降低了外植体的污染率,并且较好地诱导出愈伤组织和丛生芽;植物生长调节剂TDZ、6-BA都能诱导出愈伤组织,但是只有6-BA的培养基中没有丛生芽的发生;最适培养基为改良MS+6-BA2.0 mg/L+IAA0.02 mg/L+TDZ 0.1 mg/L+AgNO35.0 mg/L+蔗糖30 g/L+琼脂6.8g/L.其愈伤组织诱导率为90%,丛生芽诱导率为85.7%.这为后续生根研究奠定了基础.Morus notabilis Schneid, a rare and endangered Morus species, is distributed in mountainous re- gions in Yunnan and Sichuan. Being a haploid species, it is an important material for basic study of the genus Morus. The present study was intended to establish an efficient tissue culture system for plantlet regeneration from M. notabilis so as to maintain this valuable germplasm resource and do further study. The contents of the study included explant selection, medium composition optimization and investigation of the effects of different plant growth regulators on shoot regeneration. Of the explants studied, stem segments were prone to pollution, and leaves could produce calli only. With winter axillary buds as explants, pollu- tion rate was greatly reduced and calli and multiple shoots were readily induced. Of the plant growth regulators studied, TDZ and 6-BA promoted callus induction, but multiple shoots were not induced in media containing 6-BA only. The optimum medium was the modified MS supplemented with 6-BA 2.0 mg/L, IAA 0.02 mg/L, TDZ 0.1 mg/L, AgNO3 5 mg/L, sugar 30 g/L and agar 6.8 g/L, the callus induction rate and the multiple bud induction rate on it being 90% and 86%, respectively.
