摘 要:目的探讨γ干扰素(IFN-γ)诱导吉兰-巴雷综合征(GBS)患者外周血CD4+CD25-T细胞转化为CD4+CD25+调节性T细胞的可行性。方法不同浓度IFN-γ刺激GBS患者外周血中的CD4+CD25-T细胞。Real-time PCR检测诱导CD4+CD25+调节性T细胞中FoxP3的表达,共培养检测其抑制功能;结果与健康人中天然的CD4+CD25+调节性T细胞比较。结果IFN-γ可诱导CD4+CD25-T细胞转化为CD4+CD25+调节性T细胞。IFN-γ40 ng·mL-1诱导的CD4+CD25+调节性T细胞中FoxP3表达含量最高,但仍然低于天然的CD4+CD25+调节性T细胞;其抑制能力最强,与天然的CD4+CD25+调节性T细胞比较差异无统计学意义。结论 IFN-γ可诱导GBS患者外周血CD4+CD25-T细胞转化为CD4+CD25+调节性T细胞,IFN-γ40 ng·mL-1诱导的CD4+CD25+调节性T细胞表型和功能与天然的CD4+CD25+调节性T细胞相当。Aim To investigate the possibility of interferon-γ (IFN-γ) to induce CD4^+CD25^+T cells from Guillain-Barr6 syndrome (GBS) patients to CD4^+CD25^+ regulatory T cells (CD4^+CD25^+ Tregs) in vitro. Methods IFN-y with different concentrations was used to stimulate CD4^+CD25^+ T cells. The FoxP3 expression of the inducedCD4^+CD25^+ T cells in GBS patients were detected by real-time PCR. The induced CD4^+CD25^+T cells were co-cultured with autologous CD4^+CD25^+ T cells to estimate their suppressive ability to autologous CD4^+CD25^+ T cells. The results of the induced CD4^+CD25^+ T cells in GBS patients were compared with those of naturny occured CD4^+CD25^+ regulatory Tcells nTregs in healthy donors. Results CD4^+CD25^+ T cells can be induced to CD4^+CD25^+ T cells after IFN-γ stimulating in GBS patients. The FoxP3 mRNA expression of the CD4^+CD25^+ T cells induced by 40 ng.mL^-1 IFN-γ was the highest, while it was still lower than that of nTregs in healthy donors. The induced CD4^+CD25^+ T cells in GBS patients had thesuppressive function on the proliferation of autologous CD4^+CD25^+ T cells. The CD4^+CD25^+T cells induced by 40 ng.mL^-1 IFN-γ have the strongest suppressive function and the suppressive functions had no significant statistical difference between 40 ng'mL1 IFN-)' induced CD4+CD25+ T cells and nTregs of healthy donors. Conclusion IFN-7 can induce CD4^+CD25^+ T cells to CD4^+CD25^+ T cells in GBS patients. When the concentration of IFN-γ is 40 ng.mL^-1, the induced CD4^+CD25^+ T cells are similar to CD4^+CD25^+ Tregs of healthy donors phenotypically and functionally.
