摘  要:目的探讨解耦联蛋白2(UCP2)在老年大鼠免疫器官中的表达。方法健康雄性SD大鼠30只,按年龄分为幼年组、成年组和老年组,实验室适应性喂养7 d后断头处死,分别取各组大鼠脾、胸腺组织测定丙二醛(MDA)、谷胱甘肽(GSH);测定脾、胸腺指数;通过免疫组织化学观察脾、胸腺组织中UCP2蛋白的表达与分布。结果①老年组大鼠脾、胸腺组织中MDA含量显著高于幼年组和成年组(P<0.01)。②GSH含量在老年组大鼠脾组织中显著低于幼年组和成年组(P<0.01),在老年组大鼠胸腺组织中显著低于幼年组(P<0.01)。③老年组大鼠胸腺指数、脾指数均低于幼年组和成年组。④UCP2蛋白在老年组大鼠脾、胸腺实质细胞内表达较多,在幼年组、成年组大鼠脾、胸腺实质内表达较少(几乎无表达)。结论老年大鼠脾、胸腺MDA含量升高,GSH含量下降,提示老年大鼠脾、胸腺内活性氧(ROS)水平较高,并引起UCP2表达增多,UCP2可能对延缓脾、胸腺细胞的衰老和增强免疫功能起重要作用。Objective To discuss the expression of UCP'2 in the immune organs of old-aged rats. Methods Thirty healthy male SD rats were chosen and divided into three groups : young, adult and old-age groups, fed for 7 days and then beheaded. These rats' spleen and thymus tissue were taken out respectively to test the content of malondialdehyde( MDA), glutathione( GSH), spleen and thymus index and the expression and distribution of UCP2 in the spleen and thymus organisms was observed by chemical coloration. Results ①In the old aged group, the MDA content in the spleen and thymus organisms were remarkably higher than that of the young group and aduh groups (P 〈 0. 01 ). ②On the contrary, the GSH content in the old aged rats' spleen organisms was remarkably lower than that in the young group and adult group(P 〈0. 01 ), and in the old aged rats' thymus organisms, the GSH content was remarkerbly lower than that in the young group(P 〈0. 01 ). ③In the old-aged group, the spleen and thymus index were lower than that in the young group and adult group. (④The ex pression of UCP2 in the old aged rats' spleen and thymus substantial cells turned out to be much more than in the young and adult rats' sub stantial cells where the expression nearly couldn't be observed. Conclusions MDA rises while the content of GSH descends in the spleen and thymus of old-aged rats. The result suggests that the level of ( reactive oxygen species, ROS) is relatively higher and causes the increase of UCP'2 expression. UCP2 may perform important function in staving off the aging of spleen and thymus cells and improving the immune sys tem.
