摘 要:目的探讨抑癌基因Ras相关区域家族1A(RASSF1A)基因在结肠癌组织以及相应正常结肠组织中的表达并分析其表达与结肠癌临床病理因素之间的关系。方法采用免疫组织化学SP法和Western blot法检测34例结肠癌手术标本和相应的正常结肠组织中RASSF1A蛋白的表达水平,应用RT-PCR法检测RASSF1A mRNA在结肠癌和正常结肠组织中的表达情况。结果①免疫组织化学法结果:结肠癌组织中RASSF1A蛋白表达阳性率明显低于其在正常结肠组织中的表达阳性率〔35.3%(12/34)比97.1%(33/34),P<0.05〕。结肠癌组织中RASSF1A蛋白的表达与肿瘤分化程度和TNM分期均有关(P<0.05),即高、中分化及TNM分期较低(Ⅰ+Ⅱ期)者的RASSF1A蛋白表达阳性率较高(P<0.05)。②Western blot法结果:在34例结肠癌患者中RASSF1A蛋白表达水平明显低于其在相应的正常结肠组织中的表达水平〔0.316 8±0.019 6比0.914 4±0.177 6,P<0.05〕;该结果与RT-PCR法检测到的RASSF1AmRNA表达情况基本一致〔0.158 9±0.223 7和0.572 3±0.193 9,P<0.05〕。结论 RASSF1A基因的失表达可能在原发性结肠癌的发生、发展中起重要作用,检测RASSF1A的表达情况可为结肠癌的早期诊断提供帮助。Objective To detect expression of Ras association domain family IA (RASSF1A) gene in the colonic carcinoma tissue and to analyze the relationship of this expression to its clinical features. Methods Immunohistochemistry and Western blot methods were employed for detecting the RASSF1A protein expressions in 34 colonic carcinoma tissues and corresponding normal colon tissues. RT-PCR was employed for detecting RASSF1A mRNA expression. Results ① The RASSF 1A protein expression in the colonic carcinoma tissues was significantly lower than that in the normal colon tissues by using immunohistochemistry ( 35.3% (12/34) versus 97. 1% (33/34), P〈 0.05 ). There were significant relationships of RASSF1A protein expressions to the tumor differentiation and TNM stage (P〈0. 05), in other words, the positive rates of RASSF 1A protein in the moderately and well differentiated and I + II of TNM colonic carcinoma tissues were all higher (P〈 0. 05). ② The RASSFIA protein expression in the colonic carcinoma tissues was significantly lower than that in the normal colon tissues by using Western blot [0. 316 8 +0. 019 6 versus O. 914 4_+0. 177 6, P〈 0.05 ~, which wasclosetotheresultofRT-PCR[O. 1589_+O. 2237versusO. 5723_+O. 1939, P〈O. 05). Conclusions Absentexpression of RASSF 1A gene in the colonic carcinoma tissue might play an important role in tumor genesis and tumor progression, and it might become useful early detection of the colonic carcinoma.
