摘 要:目的研究重组人促红细胞生成素(recombinant human erythropoietin,rhEPO)预处理对大鼠脑缺血再灌注后脑水肿和水通道蛋白9(aquaporin 9,AQP9)表达的影响。方法成年健康SD大鼠93只,雌雄不限,采用大鼠大脑中动脉栓塞(MCAO)模型,随机分为假手术组、手术组、rhEPO组。利用干、湿重法测定脑水含量,免疫组化法测定AQP9蛋白表达,RT-PCR法测定AQP9 mRNA表达。结果干、湿重法结果显示,rhEPO预处理能够明显减轻脑缺血再灌注后脑水肿的程度(P<0.01);免疫组化和RT-PCR法结果显示,假手术组大鼠脑组织中有少量AQP9蛋白及mRNA的表达,手术组大鼠缺血侧脑组织AQP9蛋白及mRNA表达显著增加(P<0.01),rhEPO预处理能够显著降低缺血脑组织中AQP9蛋白及mRNA的表达水平(P<0.01)。结论 rhEPO保护脑缺血再灌注损伤可能与其下调脑缺血再灌注后脑组织中AQP9的表达和减轻脑水肿相关。Objective To study the effect of recombinant human erythropoietin (rhEPO) pretreatment on brain edema and AQP9 expression in cerebral ischemia-reperfusion rats. Methods 93 healthy adult SD rats were randomly divided into the sham group, ischemia-reperfusion group and rhEPO group.The ischemia-reperfusion model was established by middle cerebral artery occlusion(MCAO), the brain water content was measured using the dry-wet weight assay, the expressions of AQP9 protein and mRNA were determined by immunohistochemistry and RT-PCR respectively. Results The brain water content was higher in ischemia-reperfusion rats than in sham rats, rhEPO significantly reduced the brain water content ( P〈0.01 ) . The expression levels of AQP9 protein and mRNA in brain were increased in ischemic brain tissue than in sham group ( P〈0.01 ) , pretreatment of rhEPO markedly decreased the expression levels of AQP9 protein and mRNA ( P〈0.01 ) .Conclusion The protective effect of rhEPO on ischemia-reperfusion damage might be related to the decrease of brain edema and expression of AQP9.
