miR-194-2-192簇与MDM2在多发性骨髓瘤中的表达及临床意义

摘　　要：目的:目的:探讨miR-194-2-192簇与鼠双微体基因2(MDM2)在多发性骨髓瘤(MM)中的表达及临床意义。方法:留取MM患者骨髓标本21例,应用嵌合荧光法(SYBR?GreenⅠ)的实时荧光定量PCR(qRT-PCR)法检测miR-194-2-192簇和MDM2的表达情况,并用2-△△Ct方法进行相对定量分析。以10例健康体检者作为对照。结果:miR-194-2-192簇在MM患者中的表达显著低于对照者,而MDM2的表达显著高于对照组(均P<0.05),且均与ISS临床分期有关(均P<0.05)。miRNA-192与MDM2呈显著负相关(r=-0.353,P=0.035)。经过不同治疗后,MM患者中硼替佐米治疗者的MDM2表达水平显著低于VAD(长春新碱+阿霉素+地塞米松)治疗者(P<0.05)。结论:MDM2是miR-192的直接靶基因。miRNA-194-2-192簇低表达和MDM2高表达可能是MM发病发展的重要因子。Objective： To investigate the expression and clinical significance of miR 194 2 192 cluster and MDM2 in multiple myeloma （MM）. Method： The levels of miR-194-2-192 cluster and MDM2 were detected by quantitative RT PCR in bone marrow of 21 MM patients. Ten cases of healthy subjects were collected as controls. Result：miR 194 2 192 cluster was significantly down-regulated while MDM2 was up regulated in MM patients compared with controls （P〈0. 05） ,and the levels were associated with the clinical stage （P〈0.05）. The expres- sion of miR-192 was negatively correlated with MDM2 （r= --0. 353, P = 0. 035）. After different treatment, the level of MDM2 decreased in patients received bortezomib compared with those received VAD （P〈0.05）. Conclu- sion：MDM2 is the target of miR-192. Low expression of miR 194 2 192 cluster and over expression of MDM2 may contribute to progression of MM.
