摘　　要：目的　检测环氧化酶 2 (cyclooxygenase 2 ,COX 2 )在结直肠正常粘膜、腺瘤和癌组织中的mRNA表达 ,探讨COX 2在结直肠肿瘤发生发展中的意义。方法　以 β actin基因为参照 ,应用逆转录 多聚酶链反应 ,检测 10例腺瘤、30例癌及相应正常粘膜组织中COX 2mRNA表达。应用密度扫描技术定量分析RT PCR产物电泳带密度。结果　COX 2mRNA在 90 %的癌组织 (2 7/ 30 )中表达明显增高 ,全距 0 0 2 0～ 0 85 ,中位数 0 5 ;在正常粘膜组织中无表达或表达不明显 ,全距 0～ 0 0 74,中位数 0 ;在部分腺瘤中表达明显增高 ,全距 0～ 0 5 6 ,中位数 0 0 1。癌与正常粘膜或腺瘤间COX 2mRNA表达差异有显著性 (秩和检验 ,P <0 0 5 )。结论　COX 2mRNA表达在部分结直肠腺瘤中增高 ,在绝大多数结直肠癌中明显增高。COX 2的激活发生在结直肠肿瘤形成的较早时期 ,在结直肠肿瘤发生发展中具有重要意义。Objective To test the expression of cyclooxygenase 2 (COX 2) gene in colorectal adenomas, cancers in contrast to that of normal mucosa.[WT5”HZ] Methods [WT5”BZ] With reference to the expression of β actin gene, the expression of COX 2 mRNA was examined in colorectal adenomas from 10 patients, cancerous tissues and adjacent noncancerous mucosa from 30 patients by reverse transcription polymerase chain reaction (RT PCR). Quantitation of relative band densities was performed using densitometry scanning techniques.[WT5”HZ] Result [WT5”BZ] Strong expression of COX 2 mRNA was detected in ninety percent of cancers with range of 0 02～0 85 (median 0 5). The difference of expression of COX 2 mRNA between cancers and normal mucosa or adenomas was significant (rank sum test, P <0 05).[WT5”HZ] Conclusion [WT5”BZ] Expression of COX 2 gene elevated in most human colorectal cancers and it is increased in a subset of adenomas. Its activation is an early event and plays a role in colorectal carcinogenesis.
