摘 要:【目的】分析研发的狂犬病减毒脂质体口服冻干活疫苗的脂质体形态、粒径分布状态、疫苗抗原包封率,确定狂犬病减毒脂质体口服冻干活疫苗的制备工艺。【方法】(1)以大豆卵磷脂、胆固醇、十八胺、VE等物质为原料,采用薄膜分散法制备空白脂质体,采用电子显微镜和激光粒径分布仪测定空白脂质体的形态与粒径分布;(2)采用冷冻干燥方法将狂犬病rSRV9减毒株与空白脂质体等体积混合后冻干,制备狂犬病减毒脂质体口服冻干活疫苗;(3)采用BCA蛋白定量法检测狂犬病减毒脂质体口服冻干活疫苗的抗原包封率。【结果】制备空白脂质体形态为大单室脂质体;平均粒径为378 nm;疫苗包封率为71.19%。【结论】薄膜分散-冻干方法制备狂犬病减毒脂质体口服冻干活疫苗,抗原包封率高、性状稳定。[ Objective ] The liposome freeze -dried oral live vaccine of rabies attenuated strain was pre- pared and its morphology of liposome, distribution and encapsulation efficiency of vaccine antigen was evalua- ted for determining the preparation process of this kind of vaccine in this article. [ Method] (1) The soy leci- thin, cholesterol, octadecylamine, and vitamin E was used as raw materials, and the blank liposome was pre- pared by film dispersion, and the morphology and particle size distribution of blank liposome was evaluated by electronic microscope and laser detector MASTER2000; (2) The rabies rSRV9 strains and the blank liposome were mixed together in the equal - volume by freeze - drying method to prepare liposome freeze - dried oral live vaccine of rabies attenuated strain ; (3) The antigen encapsulation efficiency of rabies attenuated oral livevaccine was detected by BCA protein quantitative method. [ Results] The prepared blank liposome was large single chamber, and the average size of particles was about 378 nm, and the encapsulation efficiency was about 71.19%. [ Conclusion] The liposome freeze -drying oral live vaccine of rabies attenuated strain was pre- pared by film dispersion and freeze - drying method was stable and had high efficiency of antigen encapsula- tion.
