摘 要:目的探讨研究内分泌及化疗两种治疗方式对ER+乳腺癌患者中乳腺癌CD44+CD24-/low细胞亚群的影响,以期推动ER+乳腺癌的临床治疗。方法取ER+乳腺癌患者中乳腺癌细胞株MCF-7,进行体外培养,无水乙醇、雌二醇、以及雌二醇联合三苯氧胺的培养观察。随后选取雌二醇组中处于对数生长期的细胞,分别进行紫杉醇、吉西他滨以及阿霉素的培养观察,统计分析各组的分析结果。结果雌二醇培养组与联合培养组,细胞培养7 d、10 d以及15 d后,细胞中CD44+CD24-/low表现细胞所占的比例相比较或其分别与对照组比较均具有显著性差异(P<0.05)。而紫杉醇、吉西他滨以及阿霉素培养组中,细胞中CD44+CD24-/low表现细胞比例在培养1 d后与对照组比较有显著性差异(P<0.05);在培养15 d后与对照组比较无显著性差异(P>0.05)。结论内分泌以及化疗对ER+乳腺癌细胞均有重大影响,对于ER+乳腺癌根治具有重要意义,值得进一步研究探讨。Objective To explore the influence of edocrine and chemotherapy for the patients' cell subset CD44+CD24-low, and to promote ER+ breast cancer treatment. Methods The ER+ breast cancer cell line MCF-7 were cultured in vitro, with the ethanol, estradiol and estradiol combined with tamoxifen respectively; and the cell line with estradiol was cultured with the taxol and gemcitabine and adriamycin. Results After 7 d, 10 d and 15 d, the percent of the CD44+CD24-low cell in the estradiol group and joint training group exhibited the obvious difference (P〈0.05), and the significant difference was obtained when they were compared with the ones in the control group. The percent in the taxol and gemcitabine and adriamycin group was different from the control group after culturing 1 d (P 〈0.05), and was not significantly different from the control group after culturing 15 d (P 〉0.05). Conclusions Endocrine and chemotherapy in ER+ breast cancer cells have a significant impact for treatment ofER+ breast cancer; it is important and worthy of further study.
