摘 要:本研究应用差异凝胶电泳(Difference gel electrophoresis,DIGE)和质谱鉴定技术,系统分析了4种具有不同转移潜能的人肝癌细胞系的蛋白质表达差异。胶上鉴定了约1700个蛋白质斑点,经过比对后发现167个蛋白质点存在差异,经过质谱分析成功鉴定了146个蛋白质点,对应96个与肝癌转移相关的非冗余差异蛋白。对差异蛋白进行生物信息学分析,结果表明细胞组装和运动相关的蛋白在转移过程中被富集。差异蛋白前列腺脱氢酶(Hydroxyprostaglandin dehydrogenase,HPGD)首次被证明与肝癌转移正相关,具有抗凋亡和促侵袭的能力。Differential proteomics with difference gel electrophoresis (DIGE) and mass spectrometry (MS) was carried out on the four hepatocellular carcinoma (HCC) cell lines with different metastasis potential to identify HCC metastasis related proteins. Total of 1700 protein spots were found in the gel, 167 of which had quantitative differences between cell lines. Through MS identification, total of 146 differentially expressed protein spots, corresponding to 96 non-redundant proteins were found out. Bioinformatic analysis showed that most metastasis related proteins was involved in cell assembly and movement. Hydroxy-prostaglandin dehydrogenase (HPGD) was found to be positively related to HCC metastasis through inhibiting apoptosis and promoting invasion.
