摘  要:目的探讨20-羟蜕皮甾酮(20E)对四氯化碳(CCl4)肝损伤是否具有保护作用。方法将体重(20±2)g的小鼠随机分成4组,阳性对照组和20E实验组每日皮下注射2 mg·kg-1的20E注射液1次,连续7 d,正常组和模型组注射等量的生理盐水。实验d 7注射20E 2 h后,模型组和20E实验组腹腔注射剂量为10 ml·kg-1的0.25%CCl4大豆油溶液,正常组和阳性对照组注射等体积大豆油,禁食24 h后摘除小鼠眼球取血,测定血清中谷丙转氨酶(ALT),谷草转氨酶(AST)活性和血糖(Glu)的含量。结果模型组ALT和AST活性比正常组分别升高29.1%和29.5%(P﹤0.05);20E实验组较模型组二者分别降低了31.7%和39.5%(P﹤0.05);阳性对照组和20E实验组Glu浓度均比正常组减少20.3%以上(P﹤0.05),较模型组减少了21.4%以上(P﹤0.05)。结论 20E可能对CCl4肝损伤具有一定的改善作用。OBJECTIVE The study aimed to determine if 20-hydroxyecdysone (20E) is protective against carbon tetrachlo- ride (CCL)-induced liver injuries. METHODS Mice (weighing 20 :l: 2 g) were randomly divided into four groups; the pos- itive control group and 20E treatment group were subjected to a subcutaneous injection of 2 mg. kg-' of 20E per day for 7 con- secutive days, and the normal group and model group were injected with the same volumes of saline solution instead. On the 7th day of the experiment, abdominal cavities of the model group and the 20E treatment group were injected with l0 ml-kg-1 of 0.25% CCL in soybean oil 2 h after receiving the 20E injection, while the normal and positive control groups were injected with the same volume of soybean oil instead. After 24 h of fasting, blood was drawn from removed eyeballs of the mice, and the levels of alanine aminotransferase (ALT) , aspartate aminotransferase (AST) activity, and glucose (Glu) in the sera were measured. RESULTS ALT and AST activities of the model group were increased by 29.1% and 29.5%, respectively, as compared with those of the normal group (P 〈 0.05) ; in contrast, ALT and AST activities of the 20E treatment group re- duced by 31.7% and 39.5%, respectively, as compared with those of the control group (P 〈 0.05). Concentrations of Glu of the positive control group and the 20E treatment group decreased by more than 20.3% than the normal group (P 〈 0.05) and by more than 21.4% than the model group (P 〈 0.05). CONCLUSION 20E may be useful in improving liver injuries induced by CC14.
