摘 要:本文采用水提醇沉法提取小丛红景天多糖,三氯乙酸法除蛋白,蒽酮-硫酸法测定多糖含量。利用体外抗氧化体系研究小丛红景天多糖对羟自由基(·OH)、过氧化氢(H2O2)、2,2-二苯基-1-苦苯肼(DPPH·)三种自由基的清除作用,微量液体稀释法测定小丛红景天多糖的抑菌作用。结果表明,小丛红景天多糖含量为4.08%,其多糖对·OH、H2O2、DPPH·具有良好的清除能力,其清除率均随浓度的增大而增强。与Vc相比,小丛红景天多糖对·OH、H2O2、DPPH·清除能力低于Vc。体外抑菌实验证明小丛红景天多糖对大肠埃希氏菌、枯草芽胞杆菌、金黄色葡萄球菌和白假丝酵母菌四种供试菌株均有较好的抑制作用。研究结果表明,小丛红景天植物多糖在医药、保健等方面,具有较好的开发利用价值。The polysaecharide was isolated from Rhodiola dumulosa (Franeh) S. H. Fu with the method of water-dissolving and ethanol-precipitating. The concentration of polysaeeharide was determined by Anthranone-sulphurie acid method. The scavenging capacities of R. dumulosa polysaceharldes against hydroxyl free radicals ( · OH) ,hydrogen peroxide ( H202 ) and 2,2-diphenyl-2-picrylhydrazyl free radicals ( DPPH · ) were evaluated. The antimicrobial activity of R. dumulosa polysaccharides was tested using trace liquid dilution method. The results showed that the average concentration of polysaccharide of R. dumulosa was 4.08%. The scavenging capacity of polysaccharides against·OH, H2O2 and DP- PH· increased with the increasing of polysaccharides concentration. Compared with Vc,the antioxidant activity of polysaccharides was weaker. The polysaccharides had antimicrobial activity against Staphylococcus aureus, Bacillus subtillis, Escherichia coli and Candida albicans. These results demonstrated that R. dumulosa polysaccharides had potentials to be used as pharmaceuticals and/or health supplements.
