摘 要:目的 观察超顺磁性纳米榄香烯对人大肠癌细胞增殖和侵袭的影响,并探讨其可能机制.方法 制备超顺磁性纳米榄香烯,采用不同浓度的超顺磁性纳米榄香烯处理人大肠癌HCT116细胞后,以噻唑蓝(MTT)法检测癌细胞增殖,以Transwell方法检测癌细胞侵袭能力,采用实时荧光定量聚合酶链反应(FQ-PCR)方法检测癌细胞微小RNA(miR)-155 mRNA水平.结果 超顺磁性纳米榄香烯对人大肠癌HCT116细胞增殖具有一定的抑制作用.在2.5~25.0 mg/L浓度范围内,超顺磁性纳米榄香烯对癌细胞的抑制与浓度呈正相关(r =0.685);Transewell结果显示,空白对照组、不同浓度(5、10、20 mg/L)超顺磁性纳米榄香烯处理组细胞穿过滤膜的细胞分别为(44.8±1.5)、(32.2±1.3)、(18.2±1.5)和(8.6±1.2)个.FQ-PCR结果显示,与空白对照组比较,超顺磁性纳米榄香烯组miR-155 mRNA水平明显下调,且呈浓度依赖性(P<0.01).结论 超顺磁性纳米榄香烯可降低人大肠癌细胞增殖、侵袭能力,下调miR-155是其重要机制之一.Objective To study the effects and mechanism of elemene on proliferation and invasion of human colorectal cancer cell.Methods Superparamagnetic nano elemene was prepared.After human colorectal cancer HCT116 cell were treated with different doses of superparamagnetic nano elemene,and proliferation of cancer cell was studied by methyl thiazol tetrazolium (MTT) assay,and invasion ability were determined by transwell assay,and the microRNA (miR)-155 mRNA were detected by Real-time fluorescent quantitative polymerase chain reaction (FQ-PCR) assay.Results Superparamagnetic nano elemene has an inhibitory effect on the proliferation ofhuman colorectal cancer HCT116 cell line.In the concentration range from 2.5 mg/L to 25.0 mg/L,the inhibition was positively correlated with concentration of magnetic nano elemene on cancer cells (r =0.685) ; Transewell results showed that,was superparamagnetic the cells treated cells through membrane filter was 44.8 ± 1.5,32.2 ± 1.3,18.2 ± 1.5 and 8.6 ± 1.2,respectively,in the blank control group,groups of different concentrations (5,10,20 mg/L) nanometer magnetic element.FQ-PCR results showed that,compared with the blank control group,miR-155 mRNA level of superparamagnetic nano elemene group were significantly reduced,in a dose-dependent manner (P < 0.01).Conclusion Superparamagnetic nano elemene can reduce cell proliferation and invasion of colorectal cancer through down-regulating of miR-155.
