摘 要:目的 以蛋白组学方法检测早期食管癌患者与健康志愿者血浆差异蛋白质表达谱,筛选鉴定可用于食管癌早期诊断的潜在蛋白质标志物.方法 分别取Ⅰ期食管癌患者与健康志愿者空腹血浆标本各6例等量混合成2组样本,去除血浆高丰度蛋白后,利用双向电泳(2-DE)进行分离,经信息学软件分析,寻找到表达量差异在2倍以上的蛋白质,利用基质辅助激光解析电离飞行时间质谱(MALDI-TOF/TOF)技术对差异蛋白质进行鉴定.结果 Ⅰ期食管癌患者与健康人比较,筛选出表达量差异2倍以上的蛋白质点共15个,其中表达上调的有11个,表达下调的有4个,进行质谱分析后,表达上调的蛋白有α1抗凝乳蛋白酶、α2-HS-糖蛋白、富亮氨酸α2糖蛋白;表达下调的蛋白有纤维蛋白原γ链、血红蛋白α亚基.结论 Ⅰ期食管癌患者与健康人血浆的蛋白质表达谱具有一定的差异性,而鉴定出的差异蛋白则有可能成为食管癌患者早期诊断的特异性血浆肿瘤标志物.Objective To compare and analyze the expression difference of plasma proteins between early esophageal cancer and healthy individuals to search for tumor biomarkers that can be used in the early diagnosis of esophageal cancer.Methods Pooled plasma from 6 esophageal cancer patients with stage I and 6 healthy individuals as controls were collected firstly,after the removal of high-abundance plasma proteins,two-dimensional gel electrophoresis (2-DE) was used to isolate the total proteins,and then the differential protein spots with more than 2-fold were detected by 2D software,finally,the differential spots were identified by Matrix-assisted laser desorption/Ionization time of flight (MALDI-TOF/TOF) mass spectrometer.Results Fifteen differentially expressed protein spots with more than 2-fold between early esophageal cancer and healthy individuals were selected,and there were 11 up-regulated proteins and 4 down-regulated proteins among these proteins.After the identification by MALDI-TOF/TOF and analysis,the significantly up-regulated proteins were alpha-l-antichymotrypsin,alpha-2-HS-glycoprotein,leucinerich alpha-2-glycoprotein,and the significantly down-regulated proteins were fibrinogen gamma chain and hemoglobin subunit alpha.Conclusion Compared with healthy individuals,the plasma protein profile in patients with early esophageal cancer showed part of difference,and the differentially expressed protein had the potential to be the specific plasma tumor biomarkers for early diagnosis of esophageal cancer.
