类风湿关节炎患者滑膜组织中β1,4-半乳糖基转移酶-Ⅰ的表达及临床意义

摘　　要：目的检测β1,4-半乳糖基转移酶-Ⅰ(β1,4-GalT-Ⅰ)在类风湿关节炎(RA)患者膝关节滑膜组织中表达及其在RA发病过程中的作用。方法分别运用免疫组化、RT-PCR和Western blot方法检测29例炎症活动RA患者膝关节滑膜组织和7例正常膝关节滑膜组织中β1,4-GalT-Ⅰ蛋白和mRNA表达。结果 75.8%的RA患者膝关节滑膜组织中检测到β1,4-GalT-Ⅰ。与正常膝关节组织相比,β1,4-GalT-Ⅰ在RA患者滑膜组织中表达增高(P<0.01),且与显微镜下炎症严重程度(r=0.68,P<0.01)和血沉(r=0.73,P<0.01)相关。结论 RA患者膝关节滑膜组织中β1,4-GalT-Ⅰ表达水平可以作为判断病情活动的参考指标。Objective To investigate the expression and significance of β1,4-galactosyltrans- ferase-I（β1, 4-GaIT-I） in knee synovial tissues of patients with rheumatoid arthritis（RA）. Methods Using immunohistochemistry,RT-PCR and Western blot assay, the β1,4-GalT-I protein and rnRNA expressions in synovial tissues were detected in RA patients（group RA, 29 cases） and normal knee synovial tissues （group C, 7 cases）. Results The β1,4-GalT-I expressions in knee synovial tissues were detectable in 75.8% of RA patients,which were significantly higher in group RA than those in group C. The β1,4-GalT-I expressions in knee synovial tissues were closely correlated to the severity of inflammation （ r=0. 68, P〈0. 01） and erythrocyte sedimentation rate （ r=0. 73, P〈0. 01）. Conclusion β1,4-GalT-I expression level may be taken as an indicator for evaluating the severity of RA activity.
