摘  要:目的:研究双氢青蒿素(DHA)对体外培养的卵巢上皮性癌高度转移细胞株HO-8910pm黏附、侵袭能力以及体内生长的影响,并探讨其相关分子作用机制。方法:用不同浓度DHA处理HO-8910pm,采用基质胶贴壁黏附法检测DHA对癌细胞体外黏附能力的影响;用跨膜小室模型和划痕实验检测DHA对癌细胞侵袭和迁徙能力的影响。用裸鼠的HO-8910pm原位接种卵巢癌模型,检测DHA对卵巢癌侵袭和转移的影响。Western blot法检测DHA对癌细胞聚焦黏附激酶(FAK)磷酸化、MMP-2和MMP-9的体外影响。结果:(1)DHA作用于HO-8910pm细胞后,细胞增殖能力显著抑制,体外黏附和侵袭能力显著下降;(2)体内实验中,DHA可显著抑制卵巢癌腹膜转移;HO-8910pm细胞p-FAK和MMP-2蛋白水平降低,但MMP-9蛋白水平无显著降低。结论:DHA对卵巢上皮性癌细胞的增殖、黏附、侵袭能力及肿瘤转移有一定的抑制作用,其具体机制可能与抑制p-FAK、MMP-2表达水平有关。Objective :To investigate the capacity of Dihydroartiminisin (DHA) inhibiting the development and metastasis of ovarian cancer. Methods:The adhesion, invasion, and migration of human ovarian cancer cell line (HO-8910pm) was determined following DHA treatment in vitro, using Matrigel coated plate and transwell membrane chamber models, respectively. A mouse ovarian cancer model was established by orthotopic inoculation of HO-8910pm cell line in nude mice. The growth and metastasis in vivo was determined 8 weeks post-implantation in response to DHA treatment. The expression of phosphorylated focal adhesion kinase (p-FAK) and matrix metallo-proteinases (MMP-2 and MMP-9) was evaluated using Western blotting. Results:DHA inhibits ovarian cancer cell proliferation, adhesion, and invasion in vitro in a dose dependent manner, consistent with decreased expression of p-FAK and MMP-2, but not MMP-9. DHA inhibited metastasis significantly in vivo. Conclusion: DHA inhibits the pro- liferation,invasion and adhesion of HO-8910pm ovarian cancer cell line, and suppresses the metastasis in vivo.in hart via down-regulating p-FAK,MMP-2 pathway.
