摘 要:目的探讨酒精性肝病肝纤维化大鼠组织中酸性鞘磷脂酶的表达情况。方法用乙醇灌胃加高脂饮食诱导大鼠肝纤维化模型。将26只Wistar大鼠分为2组:正常对照组6只、模型组20只。用光学显微镜、电子显微镜及Masson染色观察肝组织病理学变化,应用免疫组织化学、Rail—timePCR、Westemblot等方法检测肝组织中酸洼鞘磷脂酶的表达。用SPSS软件分析,组间均数比较用Student-t检验结果肝纤维化大鼠肝脏组织中酸性鞘磷脂酶的mRNA(1.1±0.6比2.2±1.9)和蛋白质(1.00±0.36比1.80±1.26)水平均比正常组明显增加差异均有统计学意义。结论酸性鞘磷脂酶在酒精性肝纤维化发展中起到一定作用。Objective To investigate the expression of the lysosomal enzyme acid sphingomyelinase (ASMase) in alcohol-induced hepatic fibrosis using a rat model. Methods The model of liver fibrosis was induced by administration of alcohol and high fat diet using 20 rats. Six rats given no alcohol and normal diet served as the control group. Real-time PCR, western blotting, and immunohistochemistry were used to evaluate fibrosis-related changes in the mRNA and protein expressions of ASMase. Results The fibrotic liver tissues of the model rats showed significantly higher expression levels of ASMase than the non-fibrotic liver tissues of the control rats (P 〈 0.05). Conclusion Expression of ASMase is increased in the fibrotic liver tissue of an alcohol-induced hepatic fibrosis rat model, suggesting that this lysosomal enzyme may contribute to development of this disease condition.
