摘 要:目的 探讨转化生长因子β1(TGF-β1)对种植体表面成骨细胞中骨涎蛋白(BSP)表达的影响.方法 体外培养成骨细胞接种于钛片表面(模拟种植体植入后的体内环境),将其分为2个浓度刺激组和对照组.刺激组分别加入0.5、10 μg/L的TGF-β1,对照组不加TGF-β1,分别收集刺激后第2、5、7天培养的细胞,通过RT-PCR检测不同培养时间成骨细胞中的BSP mRNA表达.结果 相对半量分析结果显示:浓度为0.5 μg/L的TGF-β1可增加成骨细胞中BSP的表达(P〈0.05),而浓度为10 μg/L的TGF-β1可降低成骨细胞中BSP的表达(P〈0.05).结论 外源性TGF-β1对接种于模拟体内种植体钛片表面的成骨细胞中BSP表达的作用与其浓度有关.Objective To investigate the effects of the transforming growth factor β1 (TGF -β1 ) on the expression of bone sialoprotein in osteoblasts on implant surface. Methods Osteoblasts were seeded on the surface of Ti disc to mimic the internal environment after implantation. Then the cells were divided into two treatment groups (with 0.5 g/L or 10 g/L of TGF - β) and a control group ( without the addition of TGF -β). The cells were collected on Days 2, 5 and 7 after treatment. The expression of bone sialoprotein (BSP) mRNA was examined using RT - PCR at different times. Results According to the results of RT - PCT, the expression of BSP mRNA was increased after exposure to 0.5 g/L of TGF - β1 (P 〈 0.05), but reduced when treatment with 10 g/L of TGF - 131 (P 〈 0.05 ). Conclusion The effects of the exogenous transforming growth factor -β1 on the expression of BSP in osteoblasts on the surface of Ti disc are concen- tration - dependent.
