摘 要:筛选中华圆田螺抗菌活性物质,检测其抗肿瘤活性。本实验通过注射大肠杆菌免疫中华圆田螺,收集血淋巴并通过高温处理制成含有抗菌肽的粗提液,测定其抗菌效果和理化特性。以人体肺腺癌细胞A549为体外实验模型,通过细MTr实验,研究中华圆田螺抗菌肽对人体肺腺癌细胞A549细胞的生长抑制作用。经过分离得到含有中华圆田螺抗菌肽的粗提液。粗提液抑菌效果明显;其抑菌活性随着处理温度的升高,处理时间的延长呈现逐步降低的趋势,但仍表现出较好热稳定性;在弱酸性(pH4~6)条件下,随着酸性的降低抑菌活性呈现增加的趋势,在中性和碱性(pH7~12)条件下,随着碱性的增加抑菌活性不断降低;随着冻融次数的增加,抑菌活性略有变化;通过MTT实验表明,中华圆田螺抗菌肽对人体肺腺癌细胞A549的生长抑制作用随着浓度的增加而加强。本实验成功筛选到中华圆田螺抗菌肽,并检测其在体外对人体肺腺癌细胞A549具有抑制生长作用。Screen the antibacterial peptide from the cipangopaludina cathayensis and test its antiproliferative effects. This study extracts antibacterial peptide preliminary from cipangopaludina cathayensis and determines its antibacterial effects, physical and chemical properties. The human lung adenocareinoma cell line A549 be used as the model in vitro. MTF, mitotic index experiment and clone formation experiment be adopted to study the inhibition effect of antibiotic peptide, the bacteriostatic effect of the crude extraction is obvious; with the treating temperature increasing, treatment time extending, antimicrobial activity reduce gradually, but still show good thermal stability; Under the weak acidity (pH 4-6)conditions, antibacterial activity tended to increase as the acidity reduces. Under the neutral and alkaline (pH 7-12) conditions, antibacterial activity reduces as the alkalinity increases; With freezing and thawing times increasing, antibacterial activity changes slightly; crude extraction shows inhibition to human lung adenocarcinoma cell line A549 cells' proliferation. The antibiotic peptide from cipangopaludina cathayensis has significant inhibition effect on human lung adenocarcinoma cell line A549.
