摘 要:以中薯3号脱毒微型薯为外植体,利用根瘤农杆菌介导ODREB2B基因转化马铃薯,经PCR和RT-PCR检测有6株为转基因植株,确定ODREB2B基因已整合到马铃薯的基因组中并正常转录和翻译。在0.5%NaCl和10%PEG胁迫处理1周后,马铃薯叶片超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)活性均呈现升高的趋势,但转基因植株升高幅度大于对照,二者差异达到极显著水平(P<0.01)。结果表明,ODREB2B基因的表达提高了中薯3号的抗干旱和耐盐能力。Using the micro-tuber discs of potato variety Zhongshu No. 3 as explants, ODREB2B gene was trans- formed into Zhongshu No. 3 by Agrobacterium-mediated. Then these plants were analyzed by PCR and RT-PCR, the results showed 6 of them were transformants. We could determine that ODREB2B gene were integrated into potato genomes and transeripting and translating normally. The activities of SOD, POD, and CAT presented in- creasing trend under a week 0.5 % NaC1 and 10% PEG stress treatments. The increasing amplitude of transforma- nts were higher than the control. The difference of increasing amplitude reached to very significant level ( P 〈 0.01 ). These results demonstrated that tolerances of drought and salt of Zhongshu No. 3 were improved as a result of expression of ODREB2B gene.
