摘 要:目的 研究SMMC - 772 1细胞诱导耐药培养中P -糖蛋白变化。方法 用MTT法检测常用化疗药物对SMMC - 772 1和SMMC - 772 1/ADM细胞的毒性 ;用流式细胞仪检测SMMC - 772 1/ADM细胞P -糖蛋白表达及细胞内柔红霉素的浓度。结果 随着培养液中阿霉素浓度的递增 ,SMMC- 772 1/ADM细胞内柔红霉素的积聚减少 ,P -糖蛋白表达有逐步增加的趋势。结论 P -糖蛋白的过度表达可能是形成SMMC - 772 1细胞多药耐药的重要原因。Purpose To investigate the changes of P-glycoprotein in drug-Resistant Inducing Culture of SMMC-7721 Cells. Methods The expression of P-glycoprotein of SMMC-7721/ADM cells and the concentration of DNR in cells were studied by flow cytometric analysis .The cytotoxicity of chemotherapeutic drugs to SMMC-7721 and SMMC-7721/ADM cells were detected by MTT. Results With increasing the concentration of ADM, the accumulation of DNR in SMMC-7721/ADM cells reduced, the expression of P-glycoprotein increased.Conclusions Acquisition of multidrug resistance(MDR) is often associated with overexpression of P-glycoprotein.
